Optimized the protein A chromatography step in mAb processing platform by exploring the relationship between input parameters (DBC, elution pH, elution concentration) and responses (eluates’ pH, conductivity, yield, product quality).

Packed a 0.66* 25.8cm protein A column, wrote controlled method in Akta avant of project: Evaluation of alternate Protein A elution buffers for better control of viral inactivation pool pH.

Purified the clarified un-processing broth by Protein A. Detected eluates’ pH and Conductivity by pH/Cond. probe, detected aggregation %, HCP %, LMW % by Size-exclusion chromatography (SEC), detected mAb titer by SoloVPE and Protein A chromatography and concentrated eluates by filtration centrifugation.

Detected the molecular weight, quality and quantity of protein by Caliper GX. Analyzed the data by Labchip GX Reviewer.

Reviewed and evaluated the status and features of current nano-structure lipid carriers.

Utilized solvent evaporation technology to manufacture the PEG-DSPE and Tripalmitin/DHA based polymer-lipid hybrid nanoparticles(TPL-NPs); encapsulated the nanoparticles with small molecular chemo-drug to increase the half-life of chemo-drug from 15min to 10 hours and reduce the toxicity by enhancing the specific bio-distribution.

Encapsulated insulin into viscosity-enhanced nanocarrier (VEN) to increase the stability of its oral delivery.

Characterized the TPL-NPs by multiple analytical methods including dissolution test, encapsulation efficiency by reverse phase HPLC; size distribution and zeta-potential analysis by dynamic light scattering using Nano ZetaSizer, and morphology analysis by TEM.

Performed cell clone selection and cell culture media optimization as a part of cell line development in T-75 flasks; analyzed the cell culture parameters by Biochemical Analyzer (NOVA); monitored the cell density and viability by auto-counter and hemacytometer.

Developed a colorimetric method to determine the concentration of antimicrobial preservative (m-cresol) in α1b interferon injection; compared the results with HPLC. Greatly lowered detection cost for m-cresol by 80%.
