1. Project at Johns Hopkins Hospital, Neurology dept: (July 2010- May 2011)

Title: project which employs a novel fetal lung co-culture model to test whether HIV-infected macrophages alter biological pathways leading to uncontrolled cell growth

Designed SOP and established co-culture system for macrophages and MRC-5 cell line

Established model system to study uncontrolled pathways in MRC-5 in HIV infection.

Did 4-color flowcytometry to detect surface markers CXCR5, CD86, CD4, GFP expression.

Techniques: cell based assays,Flowcytometry, coculture systems, mammalian cell culture(triple flask culture), aseptic techniques, cell counting techniques

2. Project at Johns Hopkins Hospital, Neurology dept: (July 2010- May 2011)

Title: ‘Increased expression of Osteopontin: A Biomarker in HIV1 infection in CNS’:

Confirming the finding of increased expression of Osteopontin in HIV infected brain tissue.

Designed experiments and established pathways(cell signalling) involved in the HIV infection in brain tissue and TZMBl cells

Flowcytometry to quantify GFP signal in TZMBl,293 cells and macrophages.

Lymphocyte separation from blood samples, CD4-CD8 characterization using flowcytomtery.

Techniques: Western blot, Flowcytometry, SDS-PAGE, ELISA, Protein extraction from gels, gene cloning, RNA extraction, Microarray data analysis using IPA, Image J, Graphpad use,protein extraction, isolation, quantification using Bradford assay, sample preparation for Mass spectrometry analysis. adenovirus gene delivery for SPP gene, , transformation of competent E.coli strain with recombinant constructs, TA cloning

3. Project at Texas Tech Health Sciences Center, Lubbock: (Feb 2010- July 2010)

Title: Activation of nuclear factor 2 (transcription factor) during oxidative stress (caused by bile acid toxicity) which leads to generation of enzymes which take part in biosynthesis of antioxidants like Glutathione.

Transcription factor Nrf2 protein expression studies using western blot, SDS-PAGE

Primary cortical neuron culture experiments

Neuroblasts cell line maintenance, dose dependent and time dependent studies.

Techniques: Western blot, Protein isolation and quantification, Image J, Graphpad use.

Performed cell based assays like FACS analysis for cell viability, mammalian cell culture

4. Project at Texas Tech University, Lubbock: (Aug 2009-Dec 2009)

Title: Functional Expression, Purification, and Characterization of 3a-Hydroxysteroid Dehydrogenase/Carbonyl Reductase from Comamonas testosterone.

Performed isolation, purification, characterization of enzyme (proteins)

Enzyme kinetics study using purified enzyme from bacterial protein

Techniques: Bacterial gene cloning in vectors and its successful transformation in bacteria using competent E Coli bacteria, aseptic techniques, cell culture and plating techniques, blue-white screening, restriction enzyme digestion, primer design, PCR, RT-PCR, TA cloning, DNA-RNA extraction, affinity chromatography for protein purification, colorimetric & spectrophotometric assay, site directed mutagenesis, Mass spectrometry, trypsinization and proteomic studies of the extracted bacterial protein.

5. Project at Bangalore Biotech Labs Pvt. Ltd, Bangalore: (Dec 2006-Jan 2007)

Title: Testing Anti-proliferative Activity of Curcumine and Cytotoxic Effect of Antifoams on BHK-21 cell line:

I investigated the anti-proliferative activity of curcumine (turmeric) on immortalized cells and also the cytotoxic effect of certain specified antifoams, which are widely used in industries.

Studied growth cycle of mammalian cell lines like BHK-21 cells and CHO cells.

Performed cell based assays like MTT assay for cell viability.

Cryopreservation of cells, passaging and aseptic procedure, process development studies

Did small scale up process for CHO cells in 5 ltrs Bioreactor,optimising the parameters.
