Education
Bachelor in Biology/Microbiology 1991-1995
University of Science & Technology of Tianjin, China
Experience
Research Associate, Molecular Biology Group
Qteros, Inc. Marlborough, MA Oct. 2010-Dec.11.2011
Worked on improvement of fermentation performance by modifying metabolic enzymes.
• Generated bacterial strains with modified metabolic enzymes: Successfully generated a series of plasmids with specific integration sites using E.coli expression system. Developed a series of clostridia stain increase ethanol production.
Successfully screened the integration in targeted region.
• Conducted fermentation experiments: Executed anaerobic shaking flasks and small scale fermentations.
Research Associate
Division of Rheumatology, Immunology and Allergy
Brigham and Women’s Hospital Feb.2005-Oct.2010
• Cell cultures and co-culture: Generated and cultured cells from mice bone marrow or human cord blood; cultured dendritic cells and fibroblast cells culture, insect cells, mammalian cell culture and others; generated some cell lines; suppression co-culture mast cells and fibroblast.
• Cell line transfect ion, cytokine generation and detection: siRNA/shRNA (Mast cell proteases) cloned to Lentivirals vector and transected into LAD2 cells (gene knock out). Protease-induced cytokine generation detected by real-time RT-PCR or ELISA in endothelial cells, and fibronectin cleavage by protease deficient mast cell proteases.
• Protein generation, purification and enzymatic assays: Expressed different mast cell proteases using Baculovirus vectors in insect cells or mammalian cells. Purified recombinant proteases using anti-Flag column. Activated inactive recombinant proteases using enterokinase, performed enzymatic assays.
• Animal models: Successfully completed DSS-induced colitis (chemically induced inflammation), Derf extract-induced pulmonary inflammation, the role of mast cell proteases in angiogenesis in tumor studies.
Fermentation/bioprocess Engineer
Pharmaceutical company of Norman Bethune Medical University, China 1995-2003
• Served as Process Engineer for commercial fermentation.
• Supported fungi and microbial cell culture (media preparation and formulation) and harvest.
• Provided real time troubleshooting for unit operations (cell culture, high throughput fermentation and cGMP purification).
