1. Provincial/Ministerial Project:

Role of PVATAdrenergic Signaling in the Exercise-Induced Regulation of Vascular Circadian Rhythms Fifth Participant 2025 – Present

• Collaborated with a team of 4 members to investigate the crosstalk between perivascular adipose tissue (PVAT) and vascular circadian clocks under exercise intervention.

• Coordinated daily lab routines, including animal welfare monitoring, treadmill maintenance, and reagent inventory tracking.

• Responsible for establishing and maintaining a rhythm disorder model; performed treadmill exercise training, tissue harvesting (PVAT, aorta), and routine histological processing.

• Conducted vascular ring myography to assess endothelium-dependent and endothelium-independent vasodilation/constriction of isolated aortic rings.

• Performed RNA-seq library preparation and preliminary bioinformatics analysis (differential expression, pathway enrichment) to identify circadian-related genes modulated by PVAT adrenergic signaling.

• Validated key targets using qPCR, Western blot, and immunofluorescence staining of paraffin-embedded arterial sections. 2. Bureau-Level (City/Prefecture) Project:

Epigenetic Regulation of Smooth Muscle Cell Phenotypic Switching by PVAT-Derived Exosomal miR-122-5p in Exercise-Alleviated Obese Vascular Remodeling

Third Participant 2024 – Present

• Worked closely with 4 lab members in executing the project, ensuring smooth communication between animal surgery, cell culture, and electrophysiology subgroups.

• Managed day-to-day laboratory operations (e.g., equipment booking, buffer preparation, safety compliance) and assisted in training junior students in basic molecular techniques.

• Isolated and characterized primary vascular smooth muscle cells (VSMCs) and PVAT-derived exosomes from mouse models using ultracentrifugation and nanoparticle tracking analysis.

• Performed cell culture experiments (VSMC phenotype switching assays, exosome co-incubation) to validate the functional role of exosomal miR-122-5p.

• Conducted miRNA sequencing and bioinformatics target prediction to identify downstream pathways.

• Verified candidate genes by RT-qPCR (miRNA and mRNA) and Western blot (VSMC marker proteins).
