Prof. Hao Chen Lab, NEW JERSEY INSTITUTE OF TECHNOLOGY Mass spectrometry-based method development for analysis of emerging wastewater contaminants (PFAS)

• Designed a novel paper spray-based mass spectrometry ionization technique to simultaneously desalt and enrich samples to identify and quantify PFAS in trace levels (parts per trillion) in different matrices. Optimized spray ionization parameters, such as solvent, Spray voltage, and distance for efficient ionization, for high sensitivity. This technique is licensed to a startup company, PureTrace Labs.

Develop a quantitative mass spectrometry imaging technique using desorption electrospray ionization combined with coulometric mass spectrometry (CMS) techniques (in collaboration with Dr. Katy Margulis in the Hebrew University of Jerusalem, Israel)

• Developed a standard-free LC-HRMS technique to image and quantify the drug olanzapine and its metabolites from tissue. Separated complex mixtures of drug and its metabolites by RP-LC and optimized MS parameters to improve sensitivity, enabling quantification at fmol concentrations. This resulted in a new absolute quantification method that can quantify drug metabolites (e.g., hydroxy olanzapine) without the need for calibration standards, which are typically expensive and require time-consuming synthesis.

Standard-free absolute quantitation of post-translationally modified peptides and mAb by CMS technique

• Developed derivatization techniques to incorporate electrochemical tags to phosphopeptides and glycopeptides for absolute quantification by CMS.

• Quantified glycopeptides from tryptic mAb digest. HILIC separation was used to separate glycopeptides.

• Achieved cost-effective & standard-less absolute quantification. Probe protein 3D conformational structure

• Developed an electrochemistry-assisted mass spectrometric method to predict protein 3D structure as an alternative technique to X-ray crystallography. Labelled Lysine and Tyrosine residues of proteins with electroactive functional groups, such as Fe-C6-NHS and 4-nitrobenzene diazonium, at native pH, and quantified digested peptides to measure protein 3D structures. Compared CMS results with SASA values from X-ray crystallographic structure for method validation.

Prof. Hao Chen Lab, NEW JERSEY INSTITUTE OF TECHNOLOGY Jan-2026 to present ADC and antibody charge variant analysis using online ultrafast microdroplet digestion

• Characterized ADC drug-mimic using IdeS, PNGaseF, and Endo S2 enzymatic workflows integrated with microdroplet- accelerated digestion, followed by pseudo-DAR calculation.

• Devised a strong cation exchange-based chromatographic method for the separation of NIST mAb charge variants and performed ultrafast digestion in microseconds based on our online microdroplet digestion technique using enzymes such as IdeS and FabRICATOR Xtra.
