• Author of several successfully funded grants in conjunction with Prof. Pravindra Kumar, including DRDO, STAR-MoE, and industry-sponsored grant from THDC India Ltd.

I will use a time-resolved X-ray crystallography/CryoEM/CryoET-based structure determination approach to disseminate the mechanism of folding and misfolding of proteins in diseased and healthy states. Several genetic aberrations lead to misfolding of protein in diverse diseases like neurodegenerative diseases, type 2 diabetes mellitus, cystic fibrosis, lysosomal storage disorders, and hinder signal transduction. I will use X- ray crystallography/CryoEM to determine the structural interaction of the misfolded proteins with the other cellular proteins, which hinders the signal transduction. Further, I will use computational approaches to identify potential molecular chaperones to mediate proper folding of the misfolded proteins and will perform in vitro cell-based assays for validation. I would also like to determine the CryoEM structures of misfolded proteins in the presence and absence of chaperones to identify the structural changes mediated by the chaperones. I will also focus on animal-model studies to validate the effectiveness of the chaperons to deliver an effective treatment for the misfolded protein- mediated diseased state.

Highlights: Determined high-resolution crystal structures of phthalate plasticizers degrading enzyme EstS1 esterase in apo and in complex with different phthalate diesters, including diethyl hexyl phthalate (DEHP). Characterized its potential to degrade DEHP using biochemical and biophysical techniques (this work was covered in The Hindu News and published in the Structure Cell Press Journal

This present study highlights the potential of the thermostable and pH-tolerant EstS1 esterase from Sulfobacillus acidophilus DSM10332 to degrade high molecular weight bis(2-ethylhexyl) phthalate (DEHP) by combining biophysical and biochemical approaches along with high-resolution EstS1 crystal structures of the apo form and with bound substrates, products, and their analogs to elucidate its mechanism.

In this in-silico study, a hypothetical esterase (POB10642.1) from Sulfobacillus sp. hq2 was characterized, showing sequence identity and structural similarity with EstS1. It exhibited effective thermostability and strong binding with various phthalate diesters thus can be an effective broad spectrum phthalate diester degrading esterase.

This in-silico study revealed that natural antioxidants—sesamin, ellagic acid, capsaicin, epicatechin, and galangin—bind effectively to the catalytic site of SARS-CoV-2 Mpro, interacting with key residue Cys145.
