Isolation of genomic DNA from E.coli K12 strain

The isolated genomic DNA was subjected to PCR amplification, the amplification product was processed on a 0.7% agarose gel along with a 1 kb ladder, and the presence of SAMe was confirmed

The gene was cloned inside a pUC vector with restriction and ligation using SmaI and T4 DNA ligase

Competent cells were prepared with calcium chloride treatment and cells were transformed with recombinant pUC vector

The pUC vector was isolated and restricted with NdeI and HindIII. The restriction fragments were separated in 0.7% agarose gel.

The isolated metK gene was cloned into expression vector pET by restriction and ligation process

The vector was transformed into a competent cell for the expression of protein.

The expression was analyzed using SDS-PAGE.
