PARKINSON’S INSTITUE AND CLINIC, SUNNYVILE 2009-2012

Associate Specialist

LRRK2 is a large, widely expressed, multi-domain and multifunctional protein. LRRK2 mutations are the major cause to inherited and sporadic PD. I worked on mutant of LRRK II and some related protein, using techniques of plasmid purification, transformation, PCR.

Working on the compounds screening and characterization target to the Parkinson’s disease. The studies involve in vitro experiments using an alpha-synuclein fibrillation model, and in vivo studies in the monkey model. Biochemistry and clinical skills are used in these studies.

UNIVERSITY OF CALIFORNIA, SANTA CRUZ 2002-2007

Associate Specialist

Screening of pharmaceutical compounds to Parkinson’s disease. Looking for the inhibitors of -synuclein fibrillation for development of drugs against Parkinson’s disease. Techniques include fluorescence spectrum, mass spectrum, HPLC, electrophoresis, electronic microscopy, and mammalian cell cultures.

Protein mutagenesis: comparing kinetics of -synuclein mutant fibrillation and image of -synuclein wild type and mutant fibrils, using techniques of plasmid purification, transformation, PCR.

Conduct research for the mechanism to amyloidosis (Antibody light chain deposit diseases) and Parkinson’s disease and screening of their inhibitors. Proteins (SMA and LEN) over expression in E. coli and purification using conventional biochemistry methods (liquid chromatography, electrophoresis, extra speed centrifuge, HPLC/FPLC, et al.). Protein structures analysis using various biophysical methods.

KANSAI MEDICAL UNIVERSITY, JAPAN 2000–2001 Research fellow

Kinetics of protein folding at subzero temperature. Refolding of -lactoglobulin, ubiquitin (F45W) and other proteins/peptides at subzero temperature. Two mutants of -lactoglobulin were separated using ion-exchange chromatography. Folding kinetics as well as protein structures were studied using a stopped-flow apparatus coupled to far-UV CD, intrinsic fluorescence and SAXS (small angle x-ray scattering).

EDUCATION
