Thesis title “Studies on biofilm matrix of Vibrio cholerae”

Molecular typing of V. cholerae strains isolated from various regions of India to determine their clonal Propagation (clinical and environmental isolates).

The biofilm studies were focused on eDNA and its interaction among various component of biofilm.

Detection of eDNA release during the growth of bacteria and effects of DNase I on biofilm in time dependent manner, here we showed that eDNA is released at late log phase of bacterial growth and DNase I affects the biofilm when treated at initial time point (t=0).

As VPS is important biofilm component of V. cholerae we investigated the interaction among eDNA and VPS through various techniques, similarly we also investigated the interaction among OMVs and eDNA.

The interaction of eDNA with other components of biofilm gives strength to biofilm, here we have shown that eDNA colocalized with VPS and interacts with it. The confocal analysis showed that eDNA is present all overthe biofilm as eDNA clouds.

Physiological consequences of eDNA and OMVs on biofilm formation/colonization was also determined in mouse model with biofilm bacteria and planktonic bacteria, here we found that biofilm bacteria have more colonization as that of planktonic bacteria and OMVs increase the colonizationof bacteria as compared to eDNA. 2.
