RESEARCH THESIS
Title: Nucleolipids with Lipophilic Hydroxyacids: Activity and Properties 1. The main research project on nucleolipids focuses on their ability to form different kinds of lipid aggregates such as micelles, vesicles and lipid nanotubes which may result in nucleotide polymer in micellar/vesicle conditions. Developed a series of modified Nucleosides (A, T, U, C- analogues of ribose, de-oxy ribose, xylo sugars) and Nucleotides along with various saturated fatty acids and amino acids which were investigated for their capabilities of polymerization in micellar or in vesicular conditions.
Several lipid phosphoramidites (α- and β-hydroxy stearic acid) were prepared and conjugated to modified nucleosides to form nucleolipids and evaluated for their oligonucleotide formation under various physiological conditions. 2. Lipophilic phosphodiester prodrugs were synthesized with a small library of dideoxy and didehydro derivatives of bases-A,T, and U. Hydroxy fatty acids capability was evaluated for delivering antiviral nucleos(t)ides into cells. Lability of the phosphoester bond is investigated to monitor, either a nucleoside or a nucleotide or a cyclic nucleotide could be delivered in the cell. Out of the series of compounds synthesized, nucleolipid with d4A nuleobase has shown excellent antiviral activity against HIV virus in different cell lines. Various phosphoramidites of modified nucleosides (dideoxydidehydro nucleosides) and lipid phosphoramidites were synthesized and coupled to synthesize modified nucleolipids which were evaluated against anti-HIV RT virus. 2006-2008: Pre-Doc (M.Phil) in Organic Chemistry

, and C-nucleophiles were inserted on the calixarene skeleton by nucleophilic aromatic substitution reactions on the bis(methylsulfonyl)oxacalix[4]arene analogue. Research Project-2:
Oxacalix[2]arene[2]quinazoline macrocycles were prepared in good yields, as mixtures of syn and anti isomers, through nucleophilic aromatic substitution cyclocondensation reactions of 2,4-dichloroquinazolines and m-dihydroxybenzenes. The macrocyclization conditions were optimized and the isomeric ratio was investigated by means of one-step and fragment-coupling approaches. The oxacalixarene substitution pattern could easily be varied by altering the dichloroquinazolinyl biselectrophilic and dihydroxyaryl bisnucleophilic building blocks.
