Preparing media and reagents as needed for laboratory testing

Sterilization of media, glassware and other instruments

Perform Quality Control check

Maintain department stock cultures

Conduct microbiological assay in accordance with Standard Operating Procedures

Isolate subcultures of microbial strains from clinical samples

Projects Undertaken:

Title: Isolation of pigment producing microorganisms from soil and extraction of pigment. Abstract: Samples were collected from rhizosphere and non-rhizosphere part of soil. The pigment producing microorganisms were isolated using dilution plate technique. Isolation of pure cultures of pigment producing microorganisms were done on Yeast extract malt extract agar medium. The pigment producing microorganisms were selected and inoculated into Yeast extract malt extract broth medium and it was maintained at 37 C in a shaking incubator in the presence of light. Pigment producing cells were harvested from the fermentation broth by centrifuging and again recentrifuged in distilled water. The cells were transferred to methanol. Extracellular pigment was extracted easily. In case of intracellular pigment, the suspension containing methanol was kept in boiling water bath for a few minutes. The suspension containing methanol was dried in oven to get the dried pigment. The dry extract of pigment was dissolved in methanol to prepare a stock solution for dyeing. The fabric was immersed in the methanol extract overnight and washed in water and dried in shade.
