DESIGN AND SCALE-UP OF ADSORPTION COLUMN FOR NITRATE REMOVAL Objective: To investigate the rate of nitrate uptake by a selective resin using both empirical and theoretical kinetic models and compare the results to find the best kinetic models. Major milestones: Developed new adsorbent Tri-Ethyl Ammonium functionalized Cross-linked Chitosan Beads (TEACCB) for efficient removal of nitrate and other hazardous chemicals. Both the resin and TEACCB showed increase in nitrate removal efficiency when we increased the dosage and both the adsorbents had 100% nitrate removal efficiency even after the 10th regeneration cycle. Role or tasks completed:
Compared the characteristics of the adsorbent with commercially available synthetic resins. Conducted various batch and column adsorption studies and analyzed the effect of variable parameters such as contact time, pH, anionic concentration, adsorbent dosage, regeneration studies etc. Conducted thermodynamic study and adsorption isothermal studies. Carried out the kinetic modeling and experiments and modelling of column adsorption data. Analysed the column design and scale-up parameters. Achievements: TEACCB developed in our laboratory had good nitrate removal efficiency; it was used for the treatment of nitrate contaminated of nitrate contaminated brackish water. Benefits: From the work, it was concluded that both the laboratory synthesized biopolymeric TEACCB and commercially available resin have higher efficiency to remove nitrate from brackish water. These adsorbents can be used practically for the treatment of large scale nitrate contaminated streams. DECOLORIZATION OF DYES IN TEXTILE INDUSTRIES
Objective: To ascertain the reliability of the suitable synthetic substrates for the production of laccase enzyme from Bacillus subtilis MTCC 2414 under submerged state fermentation. The optimization, partial purification and characterization of enzyme were carried out and further applied in the degradation of synthetic dyes, such as Orange 3R, Yellow GR, and T Blue. Major milestone: The degradation of synthetic dyes, such as Orange 3R, Yellow GR, and T Blue were carried out using laccase enzyme and bacterial culture. The bacterial species which is used in the production of the laccase enzyme was Bacilus species is novel to the best of our knowledge, which is a soil bacteria and so can be easily obtained. Role or tasks completed:
Produced laccase enzyme from Bacillus subtilis MTCC 2414 under submerged fermentation. Partial purification of laccase enzyme was done by ammonium sulphate precipitation technique. Compared the degradation efficiency of synthetic dyes using microbial culture of Bacillus subtilis MTCC 2414 and free laccase enzyme. The optimization, partial purification and characterization of laccase enzyme were carried out. Achievements: The study showed the maximum dye degradation efficiency of free laccase and culture filtrate of Bacillus subtilis.
Benefits: The laccase enzyme produced by the Bacillus subtilis is thermostable and alkalophilic which has wide industrial applications, mainly in the field of dye decolourization. INDUSTRIAL TRAINING
