Physician scientist with a strong interest in genetics and septic shock of acute respiratory distress syndrome with 5 years doctoral fellow experience of basic molecular and genetics, and 5 years academic researcher experience in both in vitro and preclinical research.

Rabbit Model of Staphylococcus aureus Enterotoxin B (SEB) and SARS-CoV-2 Spike protein Toxic Shock Syndrome

Engaging in translational research aimed at developing and validating new anesthetized, mechanically ventilated rabbit model with advanced hemodynamic parameters using thermodilution catheter and echocardiography as well as longitudinal blood biomarker analyses to:

dissect mechanisms of toxin-induced acute myocardial depression with echocardiography and advanced hemodynamic monitoring with thermodilution cardiac output. This work was also funded by AstraZeneca, presented at the American Thoracic Society conference 2020 and published in Nguyen et al., Frontiers in Immunology 2023.

Ongoing research:

determine whether intravenous infusion or endobronchial instillation with two purified proteins with superantigen activities, Staphylococcus aureus Enterotoxin B (SEB) and SARS-CoV-2 Spike protein, induce toxic shock syndrome,

test whether neutralizing antibodies or active immunization confer protect against intoxication with SEB or Spike,

investigate whether SEB potentiates E. coli bacterial proliferation and lethality by inducing acute myocarditis and myocardial necrosis that serve as a substrate for subsequent ST segment abnormalities, left-ventricular wall asynchronic motion, reduced LV ejection fraction and decreased cardiac output in the rabbit model.

Rabbit Model of Klebsiella pneumoniae Blood Stream Infection

Collaborating with AiCuris Gmbh & Co. to evaluate the new treatment in combating Klebsiella pneumoniae blood stream infection using rabbit models.

ICU Rabbit Model of Pseudomonas aeruginosa Acute Pneumonia

Achieved in establishing and validating the new rabbit model of acute respiratory distress syndrome that recapitulated not only human ventilator-associated pneumonia due to Pseudomonas aeruginosa but also standardized protocols for ICU-supportive care, including fluid resuscitation, vasopressor, and humanized dosing antibiotic therapy to better mimic clinical practice to test new drugs with (1) description of the natural history of the disease; (2) PK/PD studies in uninfected and infected animals to select the human-equivalent dosing regimen of currently approved drugs; and (3) proof that the infected rabbits can be effectively treated with available antibacterial therapies.

This work was funded by FDA and published in Nguyen et al., Antimicrobial Agents and Chemotherapy 2021.

Rabbit Model of Pseudomonas aeruginosa Acute Pneumonia

Consulting ContracFect Inc., to evaluate the new treatment in combating Pseudomonas aeruginosa Acute Pneumonia using rabbit models.

Rabbit models of S. aureus as a Preclinical Drug Development Tool

Collaborate with Integrated BioTherapeutics to test a multicomponent toxoid vaccine protects against primary and secondary skin infections caused by Staphylococcus aureus in rabbit model. This work is published in Frontiers in Immunology 2021.

Collaborate with MedImmune/AstraZeneca to evidence the protective efficacy of two novel human monoclonal antibodies against staphylococcal bicomponent leucocidins in rabbit model of S. aureus pneumonia and periprosthetic joint infection. This work is published in Nguyen et al., Frontiers in Immunology 2023.

Establish an experimental ICU rabbit model to study the pathophysiology of Staphylococcus aureus septic shock to determine whether a novel immunotherapy can prevent or halt its natural disease progression. This new rabbit model of Staphylococcus aureus septic shock is used to dissect mechanisms of toxin-induced acute myocardial depression with echocardiography and advanced hemodynamic monitoring with thermodilution cardiac output,

Standardize the rabbit model of Staphylococcus aureus septic shock with fluid resuscitation and vasopressor mimicking clinical therapy to determine differences in pathophysiology of infection caused by S. aureus USA300, USA100 and USA500 and their isogenic mutant strains. This model will further be used to determine whether LukED, HlgAB/HlgCB and LukAB induce vascular failure and/or acute myocardial dysfunction. So far, the resuscitated rabbit model will be used for vaccine efficacy studies with monoclonal antibodies.
