A highly motivated and result oriented associate scientist/scientist, possessing broad R&D experience in cell biology, molecular biology, in vivo pharmacology, and immunology. Set up human progenitor CD34+ cell culture system and use it to screen and characterize hemoglobin inducers.

Establish and validate both absolute and relative qPCR methods for quantification of hemoglobin gene expression.

Validate ELISA assay for hemoglobin quantification in cell lysate samples.

Use FACS analysis to evaluate erythroid maturation and HbF hemoglobin expression.

Validate the drug target by using RNAi technology to knockdown the mRNA expression in human bone marrow CD34+ cells.

Investigate the interaction between a particular protein and DNA to promote HbF production by performing ChIP assay.

Harvest bone marrow from femurs and establish cell culture for characterization of lead compounds.

Run proliferation assay to screen and characterize cancer-killing compounds made by chemistry department.

Evaluate combination therapy with outside chemo compounds in different cancer cell lines.

Run HEMOX analyzer to screen new 3rd generation compounds in whole blood or purified hemoglobin solution.

Characterize lead compounds by sickling assay, red blood cell partition assay and hemoglobin off rate assay.

Evaluate hemoglobin’s oxygen binding affinity induced by GBT compound with measuring Bohr effect and 2,3-DPG effects.

Analyze experiment results by GraphPad Prism and present data at project meetings.

Antibody variable region DNA sequence analysis for hybridoma identification and selection.

Change antibody Fc DNA sequence and clone into expression vector for antibody isotype switching and specific function attenuation.

Master cell banking and quality control (IMPACT) for antibody producing cell lines before immunization.

Run real time PCR (qPCR) for gene expression study and target expression monitor in stable cell lines. The experiment results helped cell line selection.

Design and mutate target protein at different extracellular domains for antibody binding site identification or functional assays.

Optimize expression vector with promoter or resistance gene switching. The vector modification improved target gene expression and cell clone selection.
