Assisted studying the role of let-7 microRNA in neuronal macroautophagy (With Amy Dubinsky, Ph.D.).
oPerformed cloning and functional testing of miRNA targets using dual luciferase system, and cell culture transfection. Collected, extracted, and analyzed RNAi knockdown by qRT-PCR; quantified changes in gene expression to potential role in neuronal autophagy.
Assisted in study of antisense transcripts overlapping unstable tandem repeat regions in various neurological disorders such as Huntington’s disease, Fragile-X syndrome, and spinocerebellar ataxia type 7 and type 8. (With Paula Ladd, Ph.D.).
Techniques utilized included: mini-and maxiprep, PCR, restriction digest, agarose gel electrophoresis, qRT-PCR, cell culture, transfection, and dual luciferase assay.
