Isolation, Identification and toxigenic potentials of Mycotoxins producing Aspergillus species from foodstuffs.
Six Aspergillus species were isolated from contaminated food–stuffs viz., fruits, bread, and rice grains (CO-43, CR-1009, ADT-38, and ADT -39). Aspergillus species were grown on PDA for molecular identification. Initial identification was achieved by microscopic observation. DNA was extracted by CTAB-Method and PCR were carried out with the primers (ITS- 4 and ITS-5). PCR Products were sequenced and the sequences were BLAST in the NCBI-Database in order to indentify the Aspergillus species. Among the six- isolates three isolates belonging to Aspergillus parasiticus and three isolates belonging to Aspergillus flavus. Further assessments of mycotoxigenicity of Aspergillus species were also studied. The six isolates were grown on PDB-broth for mycotoxin production and the broth was extracted through Whattmann No.1 filter paper at different day’s intervals. Assessment of mycotoxin was analysed by HPLC. Among the isolates Aspergillus flavus produced high amount of aflatoxin on 9th day incubation. Where as low amount of aflatoixin production was observed by Aspergillus Parasiticus on 9th day incubation. Moreover in our results, demonstrated that Aspergillus flavus and Aspergillus parasiticus were commonly present in food stuffs. Incresed amount of biomass leads to increased amount of mycotoxin in different day intervals.
