Objectives
• My core competencies are drug designing (pharmaceutical), synthetic
organic and medicinal chemistry, multi-step synthesis, analytical
chemistry which especially spectroscopy liquid and gas chromatography.
Pharmacist by training, I have compelling knowledge in pharmacology
and an understanding of molecular biology. One of my important strength
is literature search, information digging and communication. I have
efficaciously managed a project and a small group of scientists in the
capacity of a project leader, extensively worked for design and synthesis
of various heterocycles and steroids.
• Demonstrated compelling understanding of the drug discovery process,
especially anti-inflammatory & anti-cancer research,
• Experienced in chemistry lab start-up & strong experience of working for
contract research & pharmaceutical companies and academic set-up,
• I have always believed in quality and ethical scientific approach for
effective drug discovery research and lead a team of young scientist and
help them find their career path.
Accomplishments
• Successfully designed, synthesized and characterized novel tri-substituted
thiazole based adenosine receptor antagonist and accomplished one of the
potent ligands for the A2A and A2B receptor sub-type.
• Successfully managed and led a kinase based project revolving around
constructing various 3-amino-1,2,4-benzotriazines, qunoxalins and the
mono and di oxides. My responsiblities apart from managing a small
group of scientist, involved scheme design, molecule design and synthesis
of about 100 compounds for a UK based pharmaceutical. My motivation
and hard work was instrumental in trouble shooting synthetic flaws with
the help of MS, NMR and LCMS and convincing the client. Thus earning
valuable business recommendation and increased the number of FTE's for
the project.
• Setting up a chemistry laboratory at the Department of Pharmacology,
Penn State University College of Medicine, I successfully designed,
synthesized and characterized triterpenoid derivatives as potential and
selective anti-melanoma compounds. The success was rewarded with
filling of 2 US patent (one granted). The most successful molecule is
placed in pre-/clinical trials.
• Actively led and managed the drug modification team which listed natural
products like taxols and other FDA-approved drugs like imatinib, sidenafil,
gemcitabine, thalidomide and pomalidomide. The modification were able to
greatly enhance either solubility, modify PK/PD, avoiding early first pass
metabolism or evading P-glycoprotein.
Experiences Research Scientist Sphaera Pharma
2/2012 - 4/2014
• Lead the Drug Modification group and manage 2 research chemists.
• Research different generally recognized as safe (GRAS) chemicals and
reagents.
• Secondly use GRAS reagents to construct drug motifs that can be
biologically cleaved and modify then suitably for their use in quaternize
(modified) drugs.
• Modification of existing FDA approved drugs, to synthesize new
derivatives that would act a pro-drugs. The aim of this research was to
modify either of the following, solubility, PK/PD profile, absorption,
obscure form first pass metabolism and P-glycoprotein 1
• The modifications also helped in effective formulations and drug re-
purposing (for other pharmacological action than it is labeled for).
• I was actively involved in design, synthesis and purification of 11beta-
hydroxy pregnenolone derivatives. The purpose of this project was to
develop steroidal compounds that closely matched the 3D molecular
structure of epichatechin. The final outcome would be activation of 5'
adenosine monophosphate-activated protein kinase to ameliorates
muscular distrophy.
Post-doctoral research scholar Penn State Hershey College of Medicine
8/2009 - 5/2011
Set-up and start a fully operation synthetic organic and medicinal chemistry
lab. Design, synthesize, elucidate and pharmacological evaluation of a
triterpenoid based anti-melanoma compounds. High-through-put screening
of compound library for synergy studies. Cell culture and cell based assays
Scientist-I Oxygen Healthcare
10/2007 - 7/2009
Lead kinase based project, design and synthesize kinase based anti-cancer
compounds for a UK based pharmaceutical company. Develop experience
in
multi-step synthesis from mg to g scale (10 mg to10g), Synthesize, purify,
and characterize intermediates and compound. Manage 3 synthetic organic
chemist at research associate level. To work with cross-functional scientific
and research teams. Conduct meetings and communication with client and
report the plan and status of the project Decisive trouble shooting of
reaction schemes & conditions.
Research Assistant Sun Pharma Advanced Research Centre
4/2002 - 5/2003
My priorities were to enhance skills in the areas of stability study, method
validation, and documentation using various analytical instruments. This
included extensive use of liquid & gas chromatography along with
dissolution apparatus. Developed methods for determination of the level of
nitrosoamines in the rubber enclosures. SOP and report preparation and
documentation.
Research Officer Zydus Cadila Healthcare
3/2001 - 4/2002
My primary responsibilities were to get rigorously trained in conducting
stability studies using dissolution apparatus and HPLC with report
generation and documentation. The stability studies were carried out on
formulations that were aimed for regulatory markets.
