Ph.D Thesis: Development novel nano medicine for drug delivery and cancer photothermal therapy
(Advisor: Dr. Huan Xie)
Developed itraconazole (ITZ)-loaded poly (lactic-co-glycolic acid) (PLGA) nanospheres
(PLGA-ITZ-NS) as a new sustained-release formulation for intravenous administration:
Desired PLGA-ITZ-NS around 200 nm was successfully developed by the
nanoprecipitation and evaporation methods, HPLC and LC-MS/MS method development
for assaying for ITZ in formulation and rat plasma, respectively. Pharmacokinetics study
showed optimal PLGA-ITZ-NS has a similar systemic bioequivalent to Sporanox while
providing a sustained plasma level.
Synthesized gold nanorods (GNRs), prepared and characterized stable GNRs/anti-carbonic
anhydrase IX (GNRs/anti-CA IX) conjugates for photothermal therapy in vitro and vivo study:
Successfully synthesized GNR with size of 30 x 10 nm, and characterized GNRs/anti-CA
IX conjugates; in vitro, binding efficacy of GNR/anti-CA IX to HT29 cells and
photothermal effect on HT29 cells; in vivo, establishing tumors in nude mice, Near
infra-red (NIR) laser photothermal ablation of tumor, which demonstrated great potential
of GNRs/anti-CA IX on localized photothermal treatment of tumor without damage of
normal tissue.
