Collaborated with pharmaceutical sponsor to investigate the molecular mechanisms involved in the body

weight regulation and improvement in glucose homeostasis after bariatric surgery in ZDF ra ts.

Optimized novel protocol to examine Insulin activity in plasma using single cell based fluorescent assay

in adipocytes

Current efforts are to Identify and characterize biomarkers for the pathophysiology of type II diabetes

using human and rodent plasma samples before and after bariatric surgery using combination of cell

based function assay, FPLC and mass spectrometry methodology. Pharmacological and molecular

biology approaches are used to characterize lead candidate proteins as novel drug targets

Characterized GIP and GPR120 receptors and its naturally occurring variants in adipocytes

Investigated the relationship between hepatic steatosis and insulin resistance by manipulating the

hepatic lipid levels in mice by either overexpression (using adenovirus) or down regulation (using

antisense oligonucleotide) of liver specific adipose differentiation related protein.

Using hyperinsulinemic-euglycemic clamp technique and other metabolic pheotyping studies our data

showed that hepatic steatosis per se does not cause hepatic insulin resistance in the a bsence of

lipogenic diet and systemic circulating factors. In contrast, our studies showed that reduction of hepatic

steatosis in high fat fed mice showed metabolic improvement in the presence of obesity.

Primary hepatocytes were used to investigate the m olecular mechanisms relating hepatic steatosis and

Insulin resistance.

Thesis: Mechanism of activation of lipolysis in adipocytes

Investigated the downstream signaling cascade in the stimulation of lipolysis using purified in vivo radio-

labeled natural lipid droplets, TG lipase and protein kinase A. Our group was the first to recapitulate

lipolysis in vitro using minimum lipolysis machinery without addition of any other cytosolic component.

The phosphorylation of 42 kDa lipid storage droplet protein 1 (LSDp1), the major phosphoprotein on the

surface of lipid droplets plays an important role in the stimulation of lipolysis.

Using enzymatic and inhibitor studies, we identified that TG lipase also has phospholipase A2 activity,

which is important for hydrolyzing phospholipid monolayer on the surface of lipid droplet during lipolysis.
