1. MS thesis -“VDUP1-dependent regulation of neuroblast proliferation and progeny differentiation in the Drosophila

CNS”

• Set up various crosses and mutants of the fruit fly Drosophila Melanogaster using gene knockdown and gene

expression techniques

• Dissecting the adult and larval fly brains and performing immunohistochemistry

• Mounting the brains and analyzing using confocal microscope and immunofluorescence

• Quantifying the gene expression or knockdown using western blots and RT-PCR

• Quantifying the expression of proteins by measuring their fluorescence intensity using CellSens Dimension

2. Molecular Cloning-Designed a new plasmid vector construct pUAST VDUP1-GFP for assessing the upregulation

of VDUP1

• Gel purified the plasmid vector using ultra clean gel spin DNA purification kit

• Cut the vector using restriction enzymes and then performed DNA ligation

• Transformation/ transfection of competent Escherichia Coli cells

• Isolated plasmid DNA using plasmid mini prep kit

• Analysis of VDUP1-GFP miniprep DNA

• Running polymerase chain reaction (PCR) to verify the VDUP1-GFP insert

• Analyzed the insert using gel electrophoresis

• The DNA was quantified and sent to Rainbow transgenics for genetic engineering of the vector into the fruit fly

Drosophila embryos thus developing a new mutant fly for further analysis for tumorigenesis
