Research focused on the function of t ranscriptional factors Foxp1 and Foxp4 on adult
hypertrophic cardiomyopathy and heart failure. Study used tamoxifen inducible Cre
mouse model conditionally knout down Foxp1 and Foxp4 genes on mouse heart, study
a lso involved adenovirus infection on primary cardiomyocytes and CHIP assay.

m ammalian cell system, purification using ultra centrifugation, gel filtration
a nd affinity binding column and H PLC methods, protein analysis and detection
by EL ISA, I EF, SDS-PAGE and western blotting, phosphatase assay.
Virology and Immunology: I solation, cultivation, purification and identification of
w ild strain vi rus, v iral identification using serological, cytopathic and
molecular biology methods, vaccine production, antibody conjugation,
i mmunofluorescence, EL ISA.
Molecular Biology: C HIP assay, cloing, PCR, qPCR, site-directed mutation, northern
b lotting, southern blotting, gene knockout and knock-in using recombineering.
Cell Biology: P rimary cell isolation and culture, cell lines culture, ES cells culture and
d ifferentiation, cell growth, proliferation, apoptosis, adhesion assay, t ransfection using
shRNA or siRNA, gene over expression or knock down using lentivi rus or adenovirus on
p rimary cells.

Research focused on the function of t ranscriptional factors GATA5 and GATA4 on mouse
embryonic heart development. Study used strict knockout method completely knocked
out GATA5 and partial GATA4 gene, found out that GATA5 regulates cardiomyocytes
cooperatively with GATA4.
Research also focused on using new techniques identifying new targets. Study used laser
capture microdissection (LCM) and microarray techniques identified Sam68, a RNA
b inding protein, regulates vent ricular compaction, gene knockout method fur ther
confi rmed this function.

Research focused on the inhibition function of 4B-BP1 during eukaryotic t ranslation
t hrough PI3K signaling pathway. Study used 2-dimentional isoelectric focusing
( IEF)/SDS-PAGE and western blotting isolated and identified a series of phosphorylated
4E-BP1 isoforms in mouse liver t issue.

Research focused on the antigen presentation function of heat shock protein 108 to
cancer and infectious diseases. Study involved protein isolation and purification from
l iver tumors by affinity and ion exchange chromatography, protein-peptide binding
assay, and peptide released and separated by reverse-HPLC chromatography.
Research focused on identification of a wild virus isolate from a chicken f lock with a
h igh morbidity and mortality. Using HA and H I test confirmed it was an avian
i n fluenza virus. Study also involved virulence identification using MDT, EID50, ICPI
and HA thermostability methods. Gene analysis further classified the virus.
