WORK EXPERIENCE
Graduate Student Researcher
Prof. Evan R. Kantrowitz's Lab - Boston, MA - January 2005 to May 2013
Explored the influence of metal on the nucleotide regulation of enzyme.
Performed extensive enzyme kinetic study with different nucleotides in the absence and presence of metals.
Determined the structure of a mutant protein and studied its significance related to the function of the protein
using X-ray crystallography. Work involved protein
expression, purification, crystallization, X-ray data collection and structure
determination of protein. Techniques and methods used include site-directed
mutagenesis, column chromatography, polyacrylamide gel electrophoresis,
hanging drop and microdialysis methods to obtain crystals, X-ray data analysis and structure refinement using
d*TREK, CNS, MOLREP in CCP4 suite, Phenix and COOT.
Utilized virtual high throughput screening to identify potential small molecules as drug candidates for protein.
Computational tools used for the structure based
drug design include AutoDock 3.0, DOCK5, GOLD, Surflex and Xscore.
Developed biochemistry assay to evaluate the inhibition of ligands against protein target.
