■ Resolved synthetic challenges in a medicinal chemistry project by developing a novel, multi-component Ugi 4CC-aza-Wittig-Intramolecular Quinazolinone rearrangement methodology for previously inaccessible and biologically active amidines.

■ Reduced time, cost and waste by 50%, and improved overall yields by 10-20% through streamlining of a 6-step, Ugi 4CC-aza-Wittig-Intramolecular Quinazolinone rearrangement sequence into a telescoped protocol for biologically relevant amidines.

■ Established structure-activity relationships (SAR) around an anti-infective sulfonamide hit compound through rational design and multi-step synthesis of 100+ analogs in less than 6 months.

■ Improved the potency by 10-folds, selectivity by 100-folds and metabolic stability by 40-folds of an anti-infective hit compound through “scaffold-hopping.”

■ Identified a highly regioselective, intermolecular Diels-Alder [4+2] cycloaddition of 6,7-indolynes via a site- selective metal-halogen exchange-elimination protocol leading to total synthesis of -cis-trikentrin B, a deceptively simple, indole alkaloid natural product.

■ Investigated the electronic and steric factors responsible for the contrasteric regioselectivity in intermolecular Diels-Alder [4+2] cycloadditions of 6,7-indolynes thus gaining mechanistic insights into the reactivity profiles of 6,7-indolyne reactive intermediates.

■ Devised and executed cheaper synthetic routes by adapting Bartoli, Fischer and Leimgruber-Batcho reactions to obtain key indole starting materials, intermediates and target molecules in >10g scale quantities.

■ Executed multi-step syntheses of unique, polycyclic benzannulated indole scaffolds and libraries via 6,7- indolyne cycloaddition and cross-coupling chemistry (e.g., Negishi, Suzuki-Miyaura and Buchwald-Hartwig). Aug 2007 - June 2009, Analyst, Analytical Solutions, Mumbai, India
