“PCR” use as a diagnostic tool for identification of beta thalassemia in north Indian population.
Babasaheb Bhimrao Ambedkar University, Lucknow
five month(January to may2016)
The present investigation was carried out to understand about beta
thalassemia which is autosomal hereditary blood disorder which reduces the
Production of hemoglobin and cause anemia. Major objective of this investigation is to diagnose beta thalassemia in north Indian population by polymerase chain reaction.in this diagnosis 1)isolated the blood DNA of by salting out method .2) DNA separated by Agarose gel electrophoresis.3) use exon 2 specific beta globin primer and amplify the DNA by polymerase chain reaction. We obtained a 632bp band which is amplified product of normal beta globin gene chain. We used an exon 2 specific primer of normal beta globin gene which is amplified the exon 2 region of beta globin chain
Because of no mutation we obtained a 632bp band which shows that there
is no mutation present in normal person.
Keywords: Beta thalassemia, DNA isolation, polymerase chain reaction.

“Extraction of secondary metabolites from fresh leaf, stem and root of Cajanus cajan (Arhar) plant.
MRD Life Sciences Pvt. Ltd., Luck now.
Six Month (From November 2016 to April, 2017).
Cajanus cajan plant collected from agriculture field, luck now for the extraction of secondary metabolites. 1) Cajanus cajan plant were taken and separated into each part. After that each part washed properly and weighing for solvent extraction method. All parts are dipped into different solvent. After that we obtained extract of each part and then we check the antibiotic activity by antibiotic sensitivity test against different pathogen.2) After that we do thin layer chromatography to check the presence of compound in each extract by using different solvent. 3)After doing this we done column chromatography to separate compound which is present in extract of leaf of Cajanus cajan plant by using 80%methanol solvent then we do 4)qualitative analysis of secondary metabolites which is shown the presence of flavonoids, carbohydrate and tannins. Then lastly we check the 5) minimal inhibitory concentration of our extract by using a B.subtilis pathogen at 620nm wavelength.
