Isolation and characterization of exopolysaccharide in Rhizobium bacteria from fenugreek

• Isolation and characterization of rhizobium bacteria- root nodule of fenugreek was used to isolate rhizobium on nitrogen free Ashby's mannitol agar plate to get pure isolated colonies.

• Further isolated colonies characteristics by their size, shape, margin, elevation, opacity, texture, pigmentation.

• Performed colony morphological characterization through gram's staining, capsule staining, motility test, biochemical test

• Studied the effect of different temperature and pH on the growth of culture and exopolysaccharide production (finding optimal growth environments)

• Detection of exopolysaccharide by acid hydrolysis method and sugar estimation by DNS method

• Structural elucidation of exopolysaccharide- dried Exopolysaccharide powder was further analyzed by NMR, FTIR, MS-MS.
