Research Experience

The CenH3 gene is a histone 3 variant found on centromeres in eukaryotes.

The function of the CenH3 protein, also called CENP-A in vertebrates is to

confer centromere identity and to serve as a foundation for the formation

of the kinetochore. The CenH3 protein consists of two domains, the N-

terminal domain and the histone fold domain (HFD). The HFD is necessary for

targeting of the CenH3 proteins to the centromeric DNA. The N-terminal tail

enables the assembly of the CenH3 protein on to the centromere and is

especially important for meiosis. These two domains have been shown to

undergo adaptive evolution in a number of species and the N-terminal tail

has been shown to undergo adaptive evolution in darters. Divergence in

CenH3 during speciation can potentially lead to incompatibility of assembly

of the CenH3 proteins from one species onto centromeric DNA of a different

species. This incompatibility could, potentially, lead to sterility of

hybrids thus forming a reproduction barrier between the species. In this

study the CENP-A gene was isolated and sequenced from seven darter species

of varying evolutionary divergence. Comparison of the sequences shows

divergence present part of the N-terminal tail among all but the most

closely related species. As such, it is possible that the adaptive

divergence observed could lead to a reproductive barrier between species

and thus potentially reinforce speciation.
