Molecular Genetics: DNA isolation, DNA cloning, DNA sequence analysis, preparation and

labeling of DNA probes, southern hybridization with non-radio labeled probes. Different

types of PCR (PCR-RFLP, PCR-RAPD, PCR-AFLP, RT-PCR). Bacterial transformation,

fungal transformation (random insertion mutagenesis by Agrobacterium tumefaciens,

protoplast mediated transformation and protoplast fusion).

Systematics: Phylogenetic analysis, raw sequence editing and polishing, alignment.

Software expertise: PAUP, McClade Phylogenetic software, MrBayes for Bayesian

Inference of Phylogeny, Model Test 3.7

DNA sequence Analysis software: Chromas, GCG Sequence Analysis Software Package,

BioEdit, FinchT and Sequencher.

Microbiology/Mycology: Gram staining, Zeil-Neelsen staining, DAPI and PI staining;

antibiotic susceptibility testing (disk diffusion, E-test strips, broth and agar dilution, MIC

determination); MB/BacT rapid culture technique. Detection and sorting of fungal nuclei

expressing nuclear fluorescent proteins FACS (Fluorescence Activated Cell Sorter).

Immunology and Serology: Bacterial cell surface protein and whole cell extract preparation;

SDS-PAGE, Commassie Brilliant Blue and silver staining; Western blot analysis; ELISA; gel

diffusion. Spectrophotometric enzyme assays; protein quantitation; serum separation from

blood; different serological and agglutination tests.
