RESEARCH AND DEVELOPMENT EXPERIENCE
Summer intern – Biopharmaceutics, Novartis Pharmaceuticals (May’09 - Present)
• Primary project – Development of a database consisting of biopharmaceutical data of Novartis-
NCEs that are/were in the clinical phase of development.
A collaborative project between Pharmaceutical Analytical Development (PHAD) and
Drug Metabolism Pharmacokinetics (DMPK) departments.
Application of GastroPlus™ (G+) and ISIS chem Draw tools to simulate
biopharmaceutical data for the drugs compiled in the database and to compare the
simulated and observed results.
Other projects
•
To develop a database of marketed pediatric formulations (drug, dosage form and age
relevant data).
Designing an optimal in vivo experiment to characterize buccal delivery of a NCE.
To support the formulation scientists in preparing the solid oral dosage forms in the
Clinical Manufacturing Unit (CMU) [hands on work with blending, granulating, drying
and compression techniques].

In vitro-in vivo correlation (IVIVC) model building and application.
GastroPlus™ Modeling to predict the human gastrointestinal absorption and IVIVC
correlations.
Formulation Development and Analytical techniques:
Size exclusion chromatography- evaluated and validated the size of LMWH.

Dynamic light scattering- analyzed the LMWH microspheres particle size.
•
Scanning electron microscopy- studied the surface morphology of microspheres.
•
Zeta potential- examined the colloidal stability and/or suspendability.
•
Determined in vitro drug release and encapsulation efficiency of microsphere formulations.
•
UV-visible spectrophotometry- colorimetric assays were used to quantify the LMWH in plasma
•
samples.
Differential scanning calorimetry- identified the LMWH compatibility with excipients.
•
Fourier transform infra red spectroscopy data evaluated and interpreted.

Spray drying- prepared microspheres using Buchi-191mini spray dryer

Capsule Coating- developed coated capsules using pan coating.

Oral and Transdermal formulations of low molecular weight heparin (LMWH)
• Oral delivery: Developed and characterized biodegradable microsphere formulations to enhance
the oral bioavailability of LMWH.
• Developed an optimized process to prepare microparticulate formulations of LMWH
containing mucolytic agents through the characterization of formulation properties such as
particle size, shape, charge and encapsulation efficiency.
• Characterization of LMWH transport across intestinal tissue using Ussing chamber and
modified Doluisio techniques.
• Assessment of oral bioavailability and pharmacokinetics of LMWH in Sprague Dawley rats.
• Transdermal systems: Development of iontophoretic, ultrasound and microneedle assisted
techniques for the transdermal delivery of LMWH.
• Determination of LMWH transport across skin tissue in in vitro studies using Franz diffusion
cells.
• Investigation of LMWH delivery across skin using passive, iontophoresis, ultrasound,
metallic and soluble maltose microneedles.
• Assessment of drug disposition and drug permeation through the skin layers using Laser
Scanning Confocal Microscopy (LSM510) studies and histological studies, respectively.
• Characterization of in vivo transdermal delivery of LMWH in male CD hairless rats
Data analysis techniques:
Non compartmental pharmacokinetic data analysis using WinNonlin® software (Version 5.2.1).

Statistical analysis and graphical representation of data (Statistix-7)®.
