Project title: The role of hipoxia and the HDAC inhibitor, VPA, as epigenetic modulators in APL

Human cell line NB4.

• I worked, for 2 years, on my research-based thesis carried out in the department of experimental and molecular pathology and oncology .This project was submitted as final examination of the MS degree in Medical Biotechnology.

• I evaluated the VPA and HIPOXIA effects on viability, histones acetylation, cell death and cell cycle.

• Human NB4 cell line (M3 acute myeloid leukemia cell line) cultures were initiated, cultured, maintained at 0.3% 02(hypoxia) and/or 20% O2 (normoxia), with or without pharmacological stimulation using VPA in different concentrations (untreated, 0.5, 1 mM).

• Cells were stained and counted with an optical microscope, lysed for western blotting assays, subjected to annexin-V and PI test and cell cycle distribution using flow cytofluorimetric assays.

• 12/2009, I took part in the 3 days of national meeting, the cell death and its physio-pathological and therapeutic significance, organized by Italian cell cultures association and stem cell foundation for human life in collaboration with University of Florence .

• Clinical and laboratory internship and traineeship, I worked for 4 years, about 1200 hours, on the professional practical training and traineeship in several diagnostic areas and clinical laboratories, 1 to 4 months in each laboratory.

• Executed and analyzed tests, of both routine and special laboratory assays of patient specimens, in many areas including cytogenetic, biochemistry, hematology, urinalysis, sierology, microbiology, pathological anatomy unit, blood bank and Trasfusional medicine unit and others to aid physicians in diagnosing and treating diseases.

• I have performed several types of diagnostic and clinical tests, manual and also automated, operated and calibrated an assortment of laboratory testing equipment and evaluated quality control within laboratory using standard laboratory test.

I collected more than 1000 blood samples, detected Hcv IgG antibodies in human serum or plasma using M.E.I.A immunoassay, and evaluated, the concentration of virus with real-time P.C.R. system COBAS® AmpliPrep/COBAS® TaqMan® Hcv Test developed by Roche diagnostic, and viral genotypes throught RIBA TEST developed by Bayer, then I analyzed statistically all collected data.
