Practical Skills:

o Molecular techniques, such as cDNA library construction, Sequencing, and analysis, Northern blot, Polymerase Chain Reaction (PCR), Expanded long template PCR, Real-Time PCR, Specific gene expression quantification and sophisticated analysis, RFLP, Dendogram analysis, sequencing, phylogenetic analysis, Low through put genotyping by Fluorescence Polarization (FP) etc.

o Illumina platform: Ultra High throughput whole genome single nucleotide polymorphism (SNP) genotyping and copy number variation (CNV) assays. Genome-wide DNA methylation assay and microarray-based gene expression by cDNA mediated annealing, selection and ligation assay.

o Data and freezer inventory maintaining for thousands of samples and reagents.

o Agilent Technologies: Huge numbers of DNA and RNA chips run on bioanalyzer.

o Cell culture in RPMI medium, PBMC separation from whole blood, stimulation and proliferation of separated PBMC, Cell breakage, ELISA, ELISPOT, immunoassays, spectroscopy, enzyme assays, protein purification, gene cloning.

o Agarose gel electrophoresis, capillary electrophoresis using Agilent Bioanalyzer 2100, DNA and RNA extraction, purification of mRNA and quality control of DNA and RNA using NanoDrop technology.

o Spectrophotometer: Single and eight- headed nano drops to measure nucleotides.

o Multiplex qPCR assay development for different intestinal parasites. Assay development for rapid diagnosis strips for multiple parasitic diseases

o Anticancer and antimicrobial drug development and treatment from the different plant extracts and chemically synthesized compound.

o Animal model experience - hamsters and mice model on leishmaniasis research.

Research Technologist August 2007 to present

o Primer and probe designing, Real-time PCR assay development and PCR standardization.

o Extraction of high quality RNA for use in downstream applications like microarray experiments to investigate the differential gene expression. Extracted RNA from different sources (e.g., Formalin Fixed Paraffin embedded archived tissues, OCT-embedded Frozen tissues, Whole blood, clot blood, separated cells etc).

o Genome-wide DNA methylation assay at nucleotide resolution and Methyl profiler assay development by real time PCR.

o Extraction of high quality DNA for use in high throughput whole genome SNP genotyping and copy number variation (CNV) analysis and the assays ran on Human 610-Quad and Human Cyto SNP12 bead chips.

o Multiplexing for cytokines and chemokines detection by Luminex bead based assay

o Whole-genome wide genotyping, Gene expression, DNA methylation assays are doing by illumina microarray platform.

Other work related soft wares – Prism, LinRegPCR, SPSS, KC junior, Xponent, Millipore Analyst, Agilent 2100, RQ manager, SDS 2.3, Chromas, Oligo, Primer3 and bead studio etc. Quick software learner as required for the data analysis.
