RESUME

EDUCATION:

B.S./M.S., Biochemistry, The Lomonosov Moscow State University (Moscow, Russia), 1977.

Graduate School at The Endelgard Institute of Molecular Biology of USSR Academy of Sciences

(Moscow, Russia), 1979-1983. Thesis adviser, Academician L. L. Kisselev.

Ph.D., Molecular Biology, thesis defense at The Institute of Molecular Biology and Genetics

(Kiev, Ukraine), 1985.

RESEARCH EXPERIENCE:

1977-1979

Junior Research Worker, Institute of Organic Chemistry (Novosibirsk, Russia).

First in the world studied the tertiary structures of tRNAs in solution. These work confirmed striking similarity with tertiary structures of crystallized tRNAs ( Valentin V. Vlassov, Anatoly T. Pusyriov, and Hans J. Gross. // Evidance from chemical modification for an unusual tertiary structure of the TC loop in rabbit liver tRNA Val1. // (1978), FEBS Letters 94, No.1, pp. 157-160.

Vlasov V.V., Puzyriov A.T., Ebel J.B., Giege R. // Chemical modification studies on the structure of tRNA Phe in solution // (1981), Bioorganic chemistry 7, No.10, pp. 1487-1495. In Russian).

1979-1983

Graduate Student, The Endelgard Institute of Molecular Biology of USSR Academy of Sciences

(Moscow, Russia). I was the second (after the American G. Attardi) in the worldwho studied the

organization of mammalian mitochondrial genome (A.T. Pusyriov, A.M. Mazo, A.G. Minchenko, and

T.A. Avdonina. // Transcriptional mapping of the rat liver mitochondrial

genome. // (1983), Gene 24, pp. 115-124).

A.M. Mazo, A.G. Mincheko, T.A. Avdonina, G.G. Gause and A.T. Pusyriov. // Discrete poly(A)-RNA species from rat liver mitochondria are fragments of 16S mitochondrial rRNA carrying its 5'-termini. // (1983), Mol. Biol. Rep. 9, pp. 155-161).

1983-1992

Research Worker, Institute of Biologically Active Substances NPO “Vector”, (Berdsk,

Novosibirsk Region, Russia).Perhaps the field of construction of normalized and subtracted

cDNA libraries is the one where I have more experience than anybody else . I even believe that I

was the first to prove the possibility of normalizing concentrations of DNA species in a mixture

[Puzyrev, A. T., Il’ichev, A. A., Petrenko, V. A. (1990). A method for equalizing concentrations

of dsDNAs in a mixture. In Abstracts of the 1st Conference on Human Genome (Moscow),

pp. 91-92, in Russian.].

1992-1993

Invited Scientist, Institute of Molecular Biology and Biotechnology, (Heraklion,

Crete, Greece). I constructed a normalized cDNA library from human erythroleukemia cells.

1993-1997

Senior Research Worker, NPO “Vector”, (Koltzovo, Novosibirsk Region, Russia) I developed diagnostics for detection of spring viremia of carp virus and hog cholera virus.

1997 – 2002

Research Assistant Professor, Division of Biological Sciences, University of Missouri, Columbia,

MO, USA. Normalized cDNA libraries showed a certain degree of normalizing but not enough to

satisfy researchers sequencing ESTs on a large scale. At the University of Missouri-Columbia in the

laboratory of Prof. Riddle (now retired) I developed an extremely effective method for construction of

cDNA libraries enriched in clones corresponding to rare mRNAs (US Patent 6,372,436). A so-called

abundant cDNA pool is used for subtraction of abundant cDNAs. The possibility of formatting such a

pool was shown in our earlier work [Il’ichev, A. A., Kischenko, G. P., Petrenko, V. A., & Puzyrev,

A. T. (1991). Mol.Biol. Engl. Tr. 25, pp. 1068-1076.]. This method decreases the concentrations of

previously abundant cDNAs to nearly undetectable and increases concentrations of previously rare

cDNAs up to 200 fold. It is probably the most effective method to date.

2006-2009

Research Associate, Ordway Research Institute, Albany, New York, USA.

I studied phenomenon of asparagine deficiency in leukemic cells.

2009- 2011

Scientist, Senex Biotechnology, Inc., Albany, New York, USA.

My task was to define a target of small artificial molecules inhibiting pathways leading to cancer. (Porter DC, Farmaki E, West DK, Schools GP, Altilia S, Chang B-D, Puzyrev AT, Lim C-U, Rokow-Kittel R, Kalurupalle S, Hurteau G, Baran PS, Wentland MP, Kiaris H, Broude EV, and Roninson IB. p21 (CDKN1A) interaction with CDK8 mediates tumor-supporting paracrine activities of damaged and senescent tumor cells and stromal fibroblasts.(2012).PNAS, accepted.

Honorary titles:

1997 Senior Research Worker in Molecular Biology

Memberships

1997-2001 Member, New York Academy of Sciences

2000-2001 Member, American Association for the Advancement of Science
