PROJECTS INVOLVED:
1. Development of Recombinant Tick vaccine in Yeast. (Ph.D. Project).
This Project involved in the cloning, expression and conventional purification of recombinant Bm95 antigen of R.microplus tick parasite. The recombinant vaccine was developed in Yeast pichia pastoris and saccharomyces cerevisiae .
2. Development of recombinant vaccine in Yeast and diagnostic tools for bovine Tuberculosis in cattle.
This Project involved in the cloning, expression and purification of different recombinant proteins for vaccine target and diagnostic tool for Mycobacterium tuberculosis in E.coli and Pichia pastoris
3. Construction of chimeric full length cDNA of a specific virus. (Novel vaccine stratergy).
This Project involved in the construction of full length viral cDNA in to expression vector. The Full length viral RNA was made by in vitro transcription and infected to cell line to recover a complete live virus.
4. Development of Yeast expressed recombinant vaccine against porcine cysticercosis in porcine and human.
This Project involved in the cloning, expression and conventional purification of recombinant TSOL18 antigen of Taenia solium parasite. This recombinant protein was expressed and secreted in to medium. The recombinant vaccine was developed in Yeast pichia pastoris.
5. Development of Yeast expressed recombinant vaccine against hookworm infections in human.
This Project involved in the cloning, expression and conventional purification of recombinant Aspartic protease antigen of human hookworm Necator Americanus. This recombinant protein was expressed and secreted in to medium. The recombinant vaccine was developed in Yeast pichia pastoris.
6. Development of VLP (Viral coat protein) based canine vaccines and designing of diagnostic kit.
This Project involved in the cloning, expression and conventional purification of recombinant VLPs exposing the immunodominant epitopes of 2 different canine viruses. The purified recombinant protein used for development of diagnostic kit.
7. Development of recombinant vaccine against enterotoxaemia in cattle.
This project involved in the development of recombinant vaccine against enterotoxaemia in E.coli for sheep.
8. Development of recombinant viral vectors.
The development of different Poxviruses as recombinant viral vectors, which carry the antigen targets and their efficiency, was analyzed in animals.
9. Analysis of Cis and Trans acting elements of AOX promoter in pichia pastoris
Investigated the regulatory mechanism of alcohol oxidase promoter in methylotrophic yeast Pichia pastoris. The project involved in the analysis of different cis acting and trans acting elements that involved in the regulation of AOX promoter in Yeast Pichia pastoris. A specific protein that binds to the AOX promoter during methanol induction was identified. The same protein was not bound to the promoter during glycerol as the carbon source. The protein was furthur charactgerized biochemically in detail.
On going project for combination Hepatitis B and Pneumococcal vaccine :
The development of combinatorial recombinant vaccine (Hepatitis B+ S.Pneumococcus) for human. The Pneumococcal epitopes will be displayed in the Hepatitis B surface antigen and the purified Hepatiits B S antigen will be analyzed for immunogenicity and efficacy against Hepatitis B infection as well as S.Pneumococcal infection.
PRODUCTS CONTRIBUTED
1. Recombinant tick vaccine:
Individually contributed to the development of recombinant tick vaccine and is ready to launch. This is first vaccine against specific tick species in the world.
2. Recombinant vaccine against cysticercosis.
Individually contributed to the development of recombinant vaccine for cysticercosis in porcine and human. The technology was already transferred to Production. All the QC and QA related documentation work has been completed.
3. Recombinant vaccine against Hookworm infections in human.
Individually contributed to the development of recombinant vaccine against hookworm infections. The project is in the final stages and technology will be transferred soon.
4. Recombinant vaccine for entero toxaemia in sheep.
Contributed and guided a group in the development of recombinant vaccine for enterotoxaemia in sheep.
EXPERTISE IN THE FIELD OF YEAST MOLECULAR BIOLOGY, E.COLI, RECOMBINANT PROTEIN EXPRESSION, PROTEIN PURIFICATION AND CHARACTERIZATION :
1. Yeast expressed recombinant TB (Mycobacterium Bovis) proteins.
More than 20 recombinant proteins of Mycobacterium bovis (E.coli expressed proteins) have been purified by affinity chromatography and the standardization at laboratory, downstream levels was completed. The purified proteins were supplied for the purpose of standardizing diagnostic kit for tuberculosis.
Around 5 recombinant proteins of Mycobacterium bovis (Pichia pastoris expressed proteins) have been purified by affinity chromatography and the standardization at laboratory, downstream levels was completed.
2. Yeast expressed recombinant tick antigen.
The recombinant tick antigen was purified from Yeast Pichia pastoris through conventional methods. The standardization at laboratory and downstream levels was completed and the technology was transferred to production unit. The vaccine has been approved by RCGM and DCGI of Govt. of India
3. Yeast pichia Pastoris expressed veterinary virus VLPs.
The purification of viral like particles from pichia pastoris, has been completed through conventional purification methods. This technology has been applied for patenting.
4. Purification of E.coli expressed plant VLPs
The conventional purification of Plant VLPs which exposes many antigenic epitopes was optimized at downstream levels and the proteins are transferred to specific labs.
5. Expression and Purification of recombinant cysticercosis antigen from pichia pastoris.
The recombinant vaccine candidate of cysticercosis was expressed as secretory form in yeast and the downstream process of purification was standardized. The technology is ready to transfer to production unit. The vaccine has been approved by RCGM of Govt. of India
6. Purification of recombinant epsilon toxin from E.coli.
The conventional purification recombinant toxin of (C.perfringens) expressed from E.coli was standardized and technology was transferred to production unit.
7. Expression and Purification of recombinant antigen for Hookworm infection from yeast.
The conventional purification of recombinant vaccine candidate for hookworm infections is partially completed. The optimization at downstream level is in progress.
8. Purification of Hepatitis B suface antigen from yeast.
The purification of Hepatits B surface antigen which displays the epitopes of S.Pneumococcus has been completed. The immunogenicity and safety studies will be initiated soon.
