BIO – FUEL PRODUCTION FROM ALGAE IN ELDORADO HOLDINGS PVT LTD, KOLKATA

To culture the algal cell (chlorella vulgaris).

Subculture from mother culture (in slant form) to broth form in conical flask for lab purpose as well as in 20 lit. pet jar for pilot plant purpose

Subculturing into slant as well as on Petri plate for preservation of culture.

Transferring of broth culture from pet jar to pilot plant and nutrients addition was done (urea& super phosphate).

Culture was left in pilot plant to multiply and after 5-7days when the colour of culture reaches to dark green, it was then harvested to collect the total biomass.

The biomass dried and dry biomass was weighted from the dry biomass the lipid content was extracted by the Sauxlet method, using butanol & n-hexane as solvent. Biomass per gram then calculated.

The algal cell concentration was measured by uv-visible spectrophotometer between 630-660nm.

The media used to culture Chlorella vulgaris was BBM (Bold’s Basal Medium),pH-6.6.

Pilot plant was a photobioreactor, imported from an European country.

After few months we switch over to pond culture method from photobioreacter, as later was not suitable in a tropical environment like Kolkata and repeat the same procedure for this pond culture technique.

JUNIOR MICROBIOLOGIST & PRODUCTION IN – CHARGE (MICROBIAL CULTURE MEDIA) IN TITAN BIOTECH LIMITED, BHIWADI, RAJASTHAN

Handling of Microscope, UV- Visible scptrophotometer, Centrifuge and other different instruments.

Experience to test the Total Viable Count (TVC) of different biological finished goods product viz., protein hydrolysate (both liquid & powder), soya protein, yeast extract powder etc.

To test the pathogen (pathogenic E. coli, Staphulococcus, Salmonella, Pseudomonas) contaminatin of different products.

To test the potency of newly produced dehydrated microbial culture media by applying the particular microorganisms (bacteria, fungi, molds etc) prescribed for that culture media– GPT Test.
