“Agrobacterium mediated transformation of AmA1 gene in Indica rice variety (HKR-46) for improving protein content”

(DBT funded, M.Sc. thesis/research topic)

• Agrobacterium strain having plasmid SB8G

• Gene used AmA1(1183 bp) (from Amaranthus hypochondriacus)

• Driven by seed specific promoter Granule Bound Starch Synthase (pGBSS)

• Neomycin phospho-transferase (npt II) as section marker

• Induction of callus from mature seeds & immature embryos (indica var. HKR-46 & japonica var. TNG-67)

• Co-cultivation of embryogenic calli with Agrobacterium tumefaciens strain (Acetosyringone)

• Selection of the transformed calli (Cefotaxime & Geneticin-418)

• Shoot & root regeneration

• DNA isolation (CTAB method)

• PCR analysis of putative transgenic plants using specific primers (F51 & R1044)

• Out of four plants of HKR-46 two plants & out of fourteen plants of TNG-67 eleven plants showed amplified

desired 1.1 kb bands

• Strategizing transformation efficiency

• Gene used super oxide dismutase (SOD)

• Induction of callus from mature seeds of VQL-2, one of the maize QPM lines (developed at VPKAS, Almora)

• Co-cultivation of maize embryogenic calli with Agrobacterium tumefaciens strain (Acetosyringone)

• Selection of the transformed calli using various antibiotic concentrations i.e., 10, 15, 25, 30 & 50 ppm
