To detect various type of diseases urine and tissues sample have been

taken. Therefore, in our study we started our work with collection of

urine and tissue samples, then isolation of proteins from urine samples,

to check the quantity of proteins, protein quantification was done for

each sample and then their quality checked by SDS-PAGE. Western blot

was performed, samples were probed with X Antibody and rest samples

was stored at 80 c for further use. RNA and DNA was isolated from

tissues and checked at nano drop for the purity of nucleic acid in sample

then run Agarose gel electrophoresis, then RNA is converted it to cDNA

for downstream and done RT-PCR and PCR of RNA and DNA for a specific

targeted gene.

Urine and tissues sample were collected from Safdarjung hospital, New

Delhi of different types of cancer patients. Among all malignant and

non-malignant cases, malignant cases like bladder, prostate, and renal

cancer, bladder cancer was observed in more during our whole study

period. Bladder cancer is predominantly found in males as compared to

females. The percentage of male patients was more reported as

compare with women.

We found good quality and quantity of DNA/RNA from tumor tissue and

protein from urine in bladder cancer patients. The validation of GATA3

in protein sample using western and dot blot suggested that these

bimolecular may be used for downstream study of bladder cancer in

future.
