TECHNICAL SKILLS

PERSONAL STRENGTH

Serum samples collected during the years 2006-2009 were tested. Hepatitis A infection was detected in 10-33% of AES cases.

Infections were high up to 6 years of age and appeared to occur throughout the year. Fecal excretion of hepatitis A virus in 12-13% and Viremia in 31% cases indicate recent infection of HAV in hospitalized AES patients.

HAV IgM positivity observed in 17-64% CSF samples suggests possibility of HAV infection as a cofactor in increasing severity of any other infection or its association in causing AES. Fecal specimens from a total of 105 children less < 5 yrs of age, hospitalized for diarrhoea were collected during Jan-March 2009.

-40.8% specimens were detected positive for rotavirus by ELISA.

-75.8% were typed for VP7 (G) and VP4 (P) genes.

-65.5% of the specimens were detected with G & P types gene

-10.3% strains remained untypable for both the genes. Each of G1P[8] and G3P[8] represented 5.2% of each of the strains, respectively while none of the specimens showed presence of a common types.

G2P[4] and G4P[8].G9P[8], G12P[6] and G12P[8] were detected at a frequency of 15.7%, 21% and 10.5%, respectively. G and P types in mixed infections represented 42.1% of the strains. Out of 342 serum samples 284 were found to be positive for anti-HAV IgM. Two hundred and twenty three of 284

(78%) specimens from different regions (Southern (n=142), Western (n=54), Eastern (n=17) and Northern (n=10) were positive for HAV RNA by RT-PCR

Nucleotide sequencing of the PCR products indicated presence of genotype IIIA. Percent nucleotide variations in Indian strains ranged from 0 -3.4 in 5’NCR, 0-8.5 in partial VP1/2A junction and 0.7-10.5 in partial RNA polymerase regions as compared to GenBank IIIA strains. Full virus particles were visualized by electron microscopy while presence of viral RNA and its 11 segments were shown by RT-PCR and PAGE respectively.

The propagation of virus was monitored up to passage 6 and was found to be infectious to normal MA104 cell culture.

Allantoic and amniotic membranes of embryonated eggs supported replication of chicken rotavirus strain Ch2. The study adds to the base line data on adaptation and propagation of avian rotaviruses in embryonated egg system.
