Project: Roles of Hsp90 in regulating epithelial mesenchymal transition (EMT) in ovarian cancer

• Produced data supporting EMT-reversal hypothesis via intracellular Hsp90 inhibition and promoter activity overexpression factors by luciferase assay.

• Optimized RNA extraction and cDNA synthesis conditions (critical genes in 2 carcinoma models by real-time PCR) and treatment conditions (cytotoxicity studies: dose and duration of 17-AAG (Hsp90 inhibitor)).

• Reproduced experiments in normal, embryonic cells as quality control measures for the experimental cell lines.

• Optimized the cell density and incubation time conditions to promote high plasmid transfection efficiency in culturing the ovarian cancer cell lines.

• Utilized and compared multiple western blot detection techniques to ensure high result accuracy of Hsp90 protein expression.

Project: CRISPR/Cas9 system knockout for pooled lentiviral library production in ovarian cancer cell lines

• Performed resistance screens on 5 target primary cell lines following successful nuclear viral infection by HEK293t mediated chemical and electroporation-based transfection techniques.

• Conducted validation experiments to confirm CRISPR-mediated gene knockdown and expression levels of Cas9-directed RNAi knockdown using quantitative Real-time PCR (qRT-PCR) in target genes.

• Optimization of pooled screening using an enhanced GFP-dCas9 fusion protein as a reporter system to indicate successful transfection.

Project: Effect of virgin coconut oil on Trimethoprim-Sulfamethoxazole toxicity in Wistar rats

• Designed dose-dependent, combination studies to determine the minimum toxicity levels that affect normal organ biochemistry.

• Performed in-depth toxicity studies on cell lysates from 20 euthanized rats specifically investigating the enzyme kinetics of liver, lungs, and testes samples using biochemical assays.

• Findings from this study indicating that virgin coconut oil reduces hepatotoxicity in Wistar rats were published in a scientific journal (Otuechere et al., 2014).
