SUMMARY OF THE PH. D WORK:
Title: Evaluation of chemopreventive potential of Diosmin on DMBA induced oral carcinogenesis - An in vitro and in vivo approach. Oral cancer is a major cause of morbidity and mortality in developing countries. Despite have advances in chemotherapy for the cancer management. However, the survival rate has not yet been improved, because poor understating about molecular mechanisms of cancer. Understanding the molecular mechanism and oncogenic signaling events would help for the prevention and/or clinical treatment of oral cancer. Dietary nutrient has been received a lot of attention in the development of novel chemotherapeutic drug. Diosmin may be a naturally occurring flavonoid glycoside that can be isolated from varied plant sources may be a potent inhibitor that functions as a strong antioxidant and anti-inflammatory, free-radical scavenging activity. In the present study, we investigated the chemopreventive efficacy of Diosmin in DMBA induced hamster buccal pouch (HBP) carcinogenesis. Oral squamous cell carcinomas (OSCC) were induced in HBPs by painting with 0.5% of 7,12- dimethylbenz(a)anthracene (DMBA), thrice in a week for 16 weeks. We observed 100 % tumor incidence by the consequence of increased levels of lipid peroxidation and decreased antioxidant and detoxification enzymes. Furthermore, defective and up-normality of inflammatory, cell proliferative, apopototic and angiogenesis events in DMBA induced hamsters.
Oral administration of Diosmin significantly inhibits tumor development as evidenced by the possible mechanism of enhanced antioxidant, detoxification enzymes and decreased lipid peroxidation, phase I enzymes and glycoconjugates in DMBA induced hamsters. Moreover, Diosmin could modulate Bax/BcL-2 ratio and significantly increased p53 expression subsequently leads to apoptosis. Furthermore, Diosmin inhibits inflammation (IL-1, IL-6, IL-8, TNF-α, COX-2, CRP), cell proliferation and angiogenesis markers (PCNA, Cyclin D1, VEGF, HIF-1α) through inhibits transcriptional activation of NF-κB, JAK/STAT-3, PI3K/AKT signaling in DMBA induced animals. Additionally, we have investigated the anticancer effect of Diosmin was evaluated in Hep-2 cell lines. Diosmin preferentially inhibited cell proliferation and induced apoptosis in Hep-2 cells through enhanced ROS generation thus in loss mitochondrial membrane potential, increased oxidative DNA damage and modulate apoptotic markers (Bcl-2, Bax, caspases-9, caspases-3). Thus, findings suggested that Diosmin have potent anti-tumor activity against DMBA induced HBP carcinogenesis. Therefore, Diosmin could be used as a novel therapeutic agent for the medical treatment and/or prevention of cancer in the future.
