Research Report on Grass Fungal Endophytes 13/02/2018 – 24/05/2018 University of Pretoria Hatfield, Pretoria

• Background research on common fungal endophytes and the nature of their interactions with host species.

• Formulation of hypothesis and questions pertaining to fungal endophytes on northern Free State grass samples.

• Given a sample of fungus from grass, and its surrounding soil, taken from the northern Free State Province- Polymerase Chain Reaction (PCR) was used to amplify a commonly used gene region for fungal systematics.

• PCR product was then sequenced, and sequence data analysed.

• This data was used for identification of the fungal species, as well as its phylogenetic data, using:

o Mothur software for amplicon sequence analysis.

o MEGA7

o Rooting and statistical testing, including bootstrapping. o BLAST search.
