Research Expertise

Research Outcomes
During M.Sc. thesis the research was done with the objectives of delineating the role of nitrogen, phosphate, metal ions and hormones in the accumulation of sucrose in the cell suspension cultures and determining the activities of enzymes of sucrose metabolism viz. sucrose synthase, sucrose phosphate and invertase in relation to sucrose accumulation in high and low sucrose synthesizing genotypes. Sugar and protein accumulation were adversely affected in sugarcane cell cultured in the presence of 6mM carbonyl cyanide m-chlorophenyl hydrazone (CCCP). The proteins declined by more than 80% while sugar by 60%. Late maturing sugarcane cultivar CoS767 was more susceptible to inhibition than early maturing cultivar C0J64.
During PhD objectives were undertaken to identify the E. coli strains from fresh water bodies that are resistant to arsenic heavy metal toxicity and to describe the divergence of the arsenic resistant determinants of arsenic -resistant bacteria. Here 17 strains exhibiting the highest levels of tolerance to the arsenic were selected and their nucleotide sequences for both arsC and ars A genes were determined. Further phylogenetic analysis was done to check the divergence in strains and the results were submitted in NCBI GeneBank with accession numbers. These genetic determinants will be of great help in constructing plant expression vectors which in turn will be useful to create transgenic plants with arsenic resistance properties for contaminated soils.
In PDFWM the project was aimed to study the role of single nucleotide polymorphisms (SNP) in migraine. For this after getting ethical clearance from Jamia Millia Islamia committee and ESIC Medical College, blood samples were collected from individuals suffering from migraine as well as healthy controls belonging to different age, race and sex and nine following SNPs along with their genes were selected to study the potentially present risk for migraine:-
rs1801133 and rs1801131, ch1, MTHFR gene
rs11172113, ch12, LRPI gene
rs4379368, ch7, Succinic HMG coenzyme A transferase (C7orf10) gene
rs10504861, ch8, MMP16 gene
rs2658199, ch1, PRDM16 gene
rs10166942, ch2, TRPM8 gene
rs2052129 and rs10156191, ch7, DAO gene
Genotyping was done by PCR-RFLP method. Genotypic and allelic frequencies were compared by SPSS 24 version. On comparing migraine subclasses, migraine with aura (MA) and without aura (MO) with control groups, the present study suggested that in MTHFR polymorphisms, the prevalence of 677CT genotype and T allele in C667T SNP influences susceptibility to MA (p < 0.05) but not to MO. Meanwhile, CC genotype in A1298C SNP could be a risk factor for migraine patients without aura (p < 0.05). For rs11172113 SNP, the CC genotype was higher in migraine (20 vs. 11%) and show significant difference on relating with CT genotype (p<0.05). This study was able to associate the role of rs 4379368 SNP with migraine susceptibility and suggested that genotype CT in rs4379368 SNP could be a genetic marker for migraine with aura (MA) but not with migraine without aura (MO). This study confirmed the association of PRDM16 to migraine susceptibility in MO and female subgroup at both genotypic and allelic level in the North Indian population and found that rs10166942 variant may be a potential marker for MA. Findings from this research work clearly indicated that a female patient with rs10156191T allele and in MA subgroup showed an increased risk for migraine. This study also confirmed that rs2052129T variant plays a significant role in migraine susceptibility in this population.
