Ph.D. Thesis Title: "Production of Lipase by Pseudomonas species"

Production of Lipase by Pseudomonas aeruginosa species was studied by using normal laboratory media and Agro-industrial wastes as substrates or nutrient sources. For this Pseudomonas aeruginosa Isolates from three different habitats Lonar Lake, Buldhana dist; Arabian sea, Juhu beach, Mumbai & Godavari river, Nanded were isolated, identified using Bergy's manual biochemical tests, using specific growth media and 16s r-RNA sequencing. Lipase production was studied at pH 9.5 (Lonar Lake isolates), pH 8.5 (Arabian Sea isolate) and pH 7 (Godavari river isolate). Assay of Lipase produced was carried out using p- Nitro phenyl acetate as substrate. Characterization of lipases was done, such as effect of incubation period, pH, temperature, carbon sources, nitrogen sources, different oil and oil industry waste substrates, metal ions, organic solvents etc. Using crude partially purified lipase. Later purification was done using techniques salt and solvent precipitation, dialysis, affinity DEAE Cellulose chromatography, SDS-PAGE and at last HPLC. The molecular wt of Lipase from Pseudomonas species L1 was found to be ~60kd. Key words: Pseudomonas species, lipase, Lonar, Arabian sea, river, SDS- PAGE
