Thesis title- N terminal proteolysis decides the ending. While the N terminal is important for GPCR signaling absence of it can deactivate the receptor and further inactivates its constitutive property. Proteolysis is important for post translational modifications like transcription, cell differentiation, cell death. There are proteases cleaving a specific site of the N terminal to activate a specific G protein coupled receptor to stimulate a signaling process for example – thrombin cleaving a specific site at protease activated receptor (PAR1) to stimulate a biological reaction known as blood coagulation cascade, on the other hand, some G protein coupled receptors don’t require proteases or peptidases to undergo proteolysis, their N terminal act as an intramolecular ligand or as an intramolecular agonist. By blocking and enhancing N terminal signaling, therapeutics can be designed for prevention of diseases caused by modulation of proteolysis at the N terminal.
