Project Title “Characterization of transglycosylases associated with cell wall biogenesis in vancomycin-resistant Staphylococcus aureus”

Objectives:

• Field sampling, isolation, and screening for Vancomycin-resistant staphylococcus aureus from bovine mastitis cow milk.

• Genotypic identification of vancomycin-resistant Staphylococcus aureus and sample preparation for MLST Sequencing.

• Study the Intracellular Staphylococcus aureus virulence in bovine mastitis cow milk using macrophages invasion assay.

• Conducted in-vitro antimicrobial susceptibility test using broth micro dilution assay moreover we also performed Mec-A gene PCR typing and Biofilm assay of vancomycin-resistant staphylococcus aureus strains.

Project Title: “Expansion and modernization of microbial Type culture collection and Gene Bank” Objectives:

• The main objectives of this project are to work on depository, to supply authentic microbial cultures and to provide related services to the scientists working in research institutions, universities, and industries.

• Regular identification of bacteria and yeast is carried out on the basis of morphological, biochemical physiological and genotypic characteristics.

• Study of Archaeal diversity from saline habitats and screening for industrially important strains using genomics approaches.

• Bacterial systematic of phototrophs and chemotrophs.

• Focused on isolation and characterization of novel antimicrobial peptides from the diversity of complex ecosystems.

Technical Expertise:

• Knowledge of microbial taxonomy, medical bacteriology, mycology, laboratory safety, aseptic technique, a handling of pathogenic bacteria, fungal isolates, culture stock preservation and maintenance. Molecular and phenotypic Species identification using Biochemical,VITEK 2,16S rRNA &18S rRNA analysis and chemo taxonomy,NCBI-BLAST,Ez-Taxon.

• In-vitro antimicrobial susceptibility testing using Broth Micro Dilution assay/PCR/Disk diffusion and VITEK 2 Test.D-Test, Vancomycin screen agar, E-strip, Performing carbapenemase detection test for the following isolates Escherichia coli, pseudomonas spp, Klebsiella spp by using carbapenemase modified hodge test,carbapenemase-imipenem-EDTA test and ESBL test.

• Biofilim assay for the Methicillin-resistant Staphylococcus aureus (MRSA strains) and other bacterial and candida species.

• Broth Micro Dilution assay of Candida, moulds dermatophytes, etc.

• Experienced in laboratory machines such as compound microscope, fluorescent microscope,MALDI-TOF and Gas chromatography, Gel filtration chromatography (G-50), HPLC, affinity chromatography, Thin layer chromatography (TLC), UV and Visible spectroscopy,PCR, Agarose Electrophoresis, protein analysis by SDS PAGES, In-Gel activity assay). Rotovap peptide extraction, AKTA pure protein purification system, Protein Estimation (BCA assay), PH meter.

• Maintaining animal cell culture like macrophage raw 264.7, J774 cell line, knowledge on Hemocytometer, and having experimental knowledge in macrophages invasion assay.
