THESIS SUMMARY

During rat spermatogenesis the chromatin undergoes dramatic remodeling; the chromatin changes from a round active state to an elongated inactive state, testis-specific linker histone H1T2 is said to play a role in the process of remodeling. It has been shown the normal chromosome structure are replaced with more basic proteins during this process. H1T2 shows a specialized localization at the “apical pole”. This is considered to be an epigenetically important area, and is hypothesized to play a role in evicting histones from the mature sperm. To understand the global function of H1T2, I studied the repertoire of the interacting protein partners of H1T2, as well as it’s genome wide occupancy. The significance of the project was to characterize H1T2, which could potentially explain the dramatic changes that occur to the nucleosome during spermatogenesis. My observation was in accordance to the existing studies which show that a different nucleosome set up is seen in the elongating rat spermatids when H1T2 is expressed. The methods used for understanding the possible role played by the linker histone protein H1T2 were, 1) Antibody validation, 2) chromatin immuno-precipitation studies 3) mass-spectrometry studies.
