Research Experience

1. Inhibitors of cell signaling pathway in T-cell lymphomas and macrophages associated

TCR engagement promotes resistance to chemotherapy in T-cell lymphomas in an ITK and GATA-3-dependent manner.

Investigated GATA-3 regulates IL-10 production and macrophage polarization in T-cell lymphomas

JAK2/FLT3 inhibitor depletes CLL-associated macrophages and impairs CLL cell viability.

Generated T-cell lymphoma mouse models using transgenic mice and xenografts.

Performed and supported projects at lab by real-time PCRs, Western blots, immunoprecipitation/Co-IP, and ELISAs;

2. Molecular mechanisms and therapeutic strategies of Alzheimer’s' disease

Recombined DNA of human myc-tagged X11α, Tau and APPswe with plasmid and cosmid vectors for transgenic animals and transformations of cell line, included enzymatic modifications, mutagenesis, transfections, expressions and immunoprecipitation.

Designated PCRs and applied for genotyping human myc-tagged X11α transgenic mice, APPswe/X11α, APPswe/Precilin1/X11α co-transgenic mice.

Performed Western blots, immunoprecipitations, RT-PCRs, ELISAs and immunochemistry stains for studying on interactions between the human X11α, Fe65 and APP in 293 cell, neuron induced from NT2 cell and transgenic animals.

Incorporated with Transgenic Animal Core, created human myc-tagged X11α (mint1) transgenic mice; Generated APPswe/X11α and APPswe/Precilin1/X11α co-transgenic mice; Constructed and screened transgenic rats of human Tau and APPswe.

3. Autoimmunity and LPLA2 (lysosomal phospholipase A2) deficiency

Genotyped and characterized LPLA2 deficiency mice, investigated into LPLA2/ leads increased autoimmunity associated with late onset Systemic Lupus Erythematosus.

Developed assays of immunofluorescence and immunochemistry for detecting IgG heavy chain, C1q and C3, and CD68 in LPLA2 / tissues.

Designated, performed and analyzed the clusters of differentiation, proliferation and apoptosis of T, B cells and macrophages in spleen, bone marrow, peripheral blood and alveolar lavage cells by flow cytometry and H3-thymidine.

Performed assays of cytokines, Anti-double-stranded DNA (dsDNA) autoantibodies,

circulating immune complexes, immunoglobulin isotopes in culture supernatant and serum.

4. Model development and therapeutics of cancer metastasis

Developed metastasis models of lymphatic and pulmonary in mouse breast cancer Ca761.

Established cell lines with different metastasis phonotypes from mouse breast cancer Ca761.

Investigated metastatic cancer organ specificities from differential metastatic models in vitro / vivo, and the interactions between metastatic cells and their host organ in molecular and cell physics.

Performed chemo resistance / sensitivity assays of metastasis cells of breast cancer by RT-PCR, MTT and H3-thymidine.
