Research Specialization in Mutagenicity, Cancer Biology and Molecular Biology

Microbiology: Expertise in bacterial molecular biology was Screening, characterization of bacteria, Genomic DNA, Plasmid DNA, and Protein extraction techniques.

Expression of Genes – Southern Hybridization, DNA sequencing- Sanger’s Di - de oxy chain termination - method.

DNA - Polymerase enzyme extracted from Thermus Aquaticus run on SDS –PAGE and Ion exchange chromatography –method was used. Taq-DNA polymerase activity was checked through PCR technique to amplify the gene of interest.

Cancer Molecular biology: At National Cancer Center, Singapore. I have been worked in two different Labs.

1. Cellular and Molecular research Lab: Different type of cell lines cultured and Molecular biology work was carried out for Cancer Gene therapy.

Experiments on Animal model have been carried out for dose fixation of drugs in order to find out the Efficacy and toxicity of drugs.

Tryphan blue assay and Haemocytometer reading was carried out. Western blot, Southern hybridization, Northern Hybridization, CDNA library construction, Mutagenesis - SDM, RFLP, AFLP and SNPs analyzed in different genes (OATP, MDR and CYT p 450- Drug metabolizing enzymes) through Automated DNA sequencer.

The following cell lines namely K562, SW480, Hep3B, HepG2, Hela, 2772, CN2, Fibroblast and Breast cancer cell lines were used in our experimental study.

2. Laboratory of Clinical Pharmacology:

Pharmacokinetics and Pharmacogenomics works have been carried out. To identify the different Exons of different genes Cytochrome p 450, OATP and MDR polymorphic effects in different populations related to ADME approach.

To understand the impact of Tamoxifen drug in different populations of Cancer patients, a study was conducted for the interaction of Drug and DNA using SNP(Single nucleotide polymorphism). SNPs were found in Cyt P 450 exons influencing the efficacy of drugs. My research paper was approved in ASCO (American Scientific Clinical Oncology) conference which was held in Florida, USA.

My Ph.D thesis was on screening of bacterium from Marine Coastal area and it’s characterization using 16s rDNA typing. Phylogenetic tree analysis result was given to NCBI (National Center for Biotechnology Information) to find out the analogues of microorganism/bacteria. The report of NCBI commented that bacterial culture developed by me was a new one even though 60% genome sequence was resembled to Pseudomonas stutzeri.

According to the result of NCBI, synthesis and characterization of the bacteria based on Silver nanoparticles made and the bacteria was named as Pseudomonas rambt-1

Drug target delivery using recent Nano Biotechnology techniques, I have done experiment in Breast Cancer cell line using silver nanoparticles to kill the cancer cells which was followed by tryphan blue assay.

In agriculture, using nanoparticles treatment for killing the pests is highly demanded to overcome the environmental hazards. Silver nanoparticles treatment is one of the approaches to kill the Paracooccus marginatus (papaya mealy bug) pest to find out the mortality rate. I got remarkable result compared with commercial pesticidal treatment.

PROJECTS ACCOMPLISHED:

PROJECT – I

Pharmacogenomics study: Personalised medicine (DRUG - DNA interaction).

SNPs Studies on Cyt-P450, MDR and OATPs families and its pharmacological effects found in different Populations of Cancer Patients (Malaysian, Chinese and Indians) samples using by PCR, RFLP, RT-PCR and DNA sequencing.

PROJECT – II

Cancer Genomics - Aurora kinase (Mitotic spindle regulator - molecular study).

AIK Gene Regulation pathways elucidates in different Cancer cell lines were HeLa, MCF-7, Hep3B, SW480, HepG2, CN-47, 2772, HEK 293 and K562 and Cancer tissues. Molecular analysis of AIK gene Regulation found using by DNA Transfection, Transformation, Southern Hybridisation, Northern hybridization, Western/immunoblot, RT-PCR and DNA Sequencing are employed.

PROJECT -III

Protein purification: Taq Polymerase production at large scale.

Thermus aquaticus/E.Coli culture was grown in 100 Lit, Bio reactor followed by Taq DNA polymerase Enzyme purified by Ion Exchange column chromatography.

PROJECT –IV

Genographic Project: (Migration of Human Genes).

To study the Migration of human genes around the World from east Africa to other parts of the world .There were 10 Lakhs number of Blood samples collected from different races and analyse by PCR followed by DNA Sequencing to find out the Gene marker.
