Worked on vascular remodeling with cPLA2 (cytosolic phospholipase A2) in human and rat smooth muscle cells. Successfully created a new double mutant phenotype by disrupting the Erg 6 gene for the ncr1 mutant, which is responsible for Niemann Pick disease in humans. In yeast, the function of this gene is not known. However, there are speculations that this gene might be involved in sterol trafficking, as confirmed by GC and GC-Mass Spectrometer. Also developed a double mutant phenotype for the arv1 gene (novel gene), which is a mediator of sterol trafficking with a functional human homolog.

Worked on transcriptional regulation of 3 C-4 demethylation genes in Saccharomyces and model studies on ncr1 and arv1 genes in Saccharomyces. Studied the roles of Erg 25, 26 and 27 genes in the removal of two methyl groups at C-4 of the sterol molecule of sterol biosynthetic pathway in saccharomyces cervisiae. The demethylation at the C-14 position appears to be the sole target of azole antifungal activities. The main objective was to analyze the 3 C-4 demethylase genes in Saccharomyces to determine the regulatory mechanisms governing these genes and to assess the possibilities of their suppression which may contribute to their resistance to antifungal agents. Achieved a very good -gal activity for the Erg 26 & 27 promoters fused to Lac Z. Further planned study the interactions between Erg 27, 26 and 25 with Erg 7 gene at the transcriptional level.

Worked on sucrose synthase gene isolation and characterization in Arabidopsis thaliana, a plant model system. This enzyme is one of the key enzymes responsible for sucrose and carbohydrate metabolism in higher plants. Only 2 genes had been reported previously. Identified 4 genes, including the 2 reported in Arabidopsis. Successfully cloned full-length cDNAs and confirmed for intron splicing and mutations by sequencing the clones.

Post Doctoral Fellow

Worked on isolation, cloning and over expression of citrate synthase gene in Arabiodopsis thaliana. The over expression of enzyme citrate synthase leads to uptake of phosphate ions via roots from soil, the excess enzyme emerging from roots acts as a chelating agent. The candidate gene was successfully isolated, sequenced and cloned into a binary vector for plant transformation. Standardized the enzyme assay for this gene. Screened Arabidopsis plants for female gametophytic mutants using transposon mutagenesized lines of Arabidopsis by gene trap and enhancer trap lines.

PhD Dissertation

(1997)

Title: ‘Induction of haploidy by anther culture and microspore culture in Pigeon pea (Cajanus Cajan (L) Mill Sp.)’ a grain legume of tropics and subtropics.

Research focused on haploidy: anther and microspore (which provide excellent experimental system for genetic transformation, mutant cell selection, haploid breeding, study of embryo development at the cellular, molecular & biochemical levels) cultures were employed for the production of haploids from pollen. Haploid chromosome number was confirmed by various experimental methodologies. Factors for callus induction, somatic embryogenesis and regeneration studies from different explants of 2 cultivars and 2 mutants were standardized.
