Summary of my Ph.D. Work
My Doctoral programme is concerned with “Influence of insecticides alone and in combination with fungicides on microflora of groundnut (Arachis hypogaea L.) soils”. Introduction of high-yielding varieties and monocroping of groundnut (Arachis hypogaea L.) in the semi-arid region of India has necessitated the use of enormously large quantities of insecticides alone and in combination with fungicides to control the major insects and pathogenic fungi of this crop. Thus organophosphorus insecticides, particularly monocrotophos and chlorpyrifos, and fungicides, mainly mancozeb and carbendazim have been widely used to combat several insect pests and pathosgenic fungi of groundnut raised in Anantapur District of Andhra Pradesh in India. This situation of extensive and intensive use of several insecticides and fungicides on groundnut would lead to environmental contamination and possible health hazards. Since, a major portion (nearly 99%) of insecticides and fungicides applied to the crop ultimately reaches the soil. These agrochemicals may act upon a wide array of non-target soil microorganisms which are of much importance to the soil fertility. The present study concerned with Influence of pesticides alone and in combinations on microflora in soils is, therefore, aimed at determining the (a) effect of monocrotophos, chlorpyrifos alone and combination with mancozeb and carbendazim on soil population of non-target microorganisms, important to the soil fertility, like nitrifiers and Azospirillum sp., bacteria, fungi and also on the elemental transformations such as ammonification, nitrification; (b) effect of the selected insecticides alone and in combination with fungicides on activities of various important soil enzymes, viz., arylamidase, dehydrogenase, myrosinase, phoshatase, protease and urease; and (c) degradation of organophosphate insecticide, monocrotophos by pure cultures of Rhodocococcus ruber strain KNUC9051 and Rhodococcus phenolicus strain DSM 44812T isolated from soil by enrichment culture technique.
The selected pesticides monocrotophos, chlorpyrifos alone and in combination with mancozeb and carbendazim, significantly enhanced the population of Azospirillum sp. and nitrifiers at the level of 2.5 to 5.0 kg ha-1 in both soils. Application of pesticides, singly and in combination, significantly enhanced the rate of ammonification at 2.5 or 5.0 kg ha-1, whereas increase in the concentration of pesticides up to 10.0 kg ha-1 resulted a slight decrease in the mineralization of peptone nitrogen in both black and red soils. Nitrification was increased in all individual and binary mixtures of pesticides treated soils up to 5.0 kg ha-1 than the controls in four weeks incubated soil samples. The rate of nitrification continued up to four weeks and then gradually decreased further with the increase of incubation period. The bacterial and fungal populations were significantly increased in soil samples received the pesticides alone and in combination at the concentration of 2.5 to 5.0 kg ha-1. The activities of arylamidase, dehydrogenase, myrosinase, phosphatase, protease and urease were increased in all in individual and binary mixtures of pesticides
