Visible Dye Based Staining For Detection Of DNA In Agarose Gel:-

In conclusion the procedures of DNA staining in electrophoresed gels can be summarized as follows:-

Methylene blue as single component stain (0.005 mg/ml) can be projected as the best when a quick response (within 30min) is needed, but has less sensitivity towards low concentrations of DNA.

The most useful gels were themselves lightly coloured at the range 0.001 to 0.005 mg/ml since it was not requiring detaining protocol.

Out of the all combinations tested, Methylene blue + Carbolfuschin (0.005 +0.001 mg/ml) and Crystal violet + Nile blue (0.005 +0.025 mg/ml) showed good results.

A formulation of Nile blue + Methylene blue + Methyl orange (0.025 +0.05 + 0.005 mg/ml) can be projected as an efficient stain since it shows good resolution of DNA band with StepUpTM 500bp ladder within one hour.
