Experiment Techniques Experience
Isolated Alkaline Phosphatase (AP) from a mutant strain of E. coli. Anion exchange chromatography was performed; fractions of the enzyme were gathered in test tube samples.
A BCIP spot test was used to determine which samples contained AP activity.
Absorbance was determined via spectrophotometry, with known concentrations and kinetic constants were able to be derived.
Compared CYT-B samples of shrews to figure out what species it belonged to and build a phylogenetic tree. Used PCR reaction, purification, performed gel electrophoresis and sequencing.
